Objective:To observe the effect of Moxibustion with Seed-Size Moxa Cone on myocardial ischemia-reperfusion injury at different stages,and to understand the correlation between this effect with Beclinl expression.Methods:This experiment is divided into two parts.Part1:Forty-two rats were randomly divided into model group,1-day moxibustion group,2-day moxibustion group,3-day moxibustion group,4-day moxibustion group,5-day moxibustion group and 7-day moxibustion group.The model of myocardial ischemia-reperfusion was established by ligating the left anterior descending coronary artery(LAD).The electrocardiogram(ECG)was monitored and the heart was taken at 4h after reperfusion,to evaluate the effect of different time pretreatment.Part2:After definite intervention time of wheat moxibustion pretreatment,sham-operated group,model group,moxibustion group and autophagy inhibitor(3-MA)group were added.6 rats respectively for ischemic 30min and reperfusion for 240min,serum cTnT levels were detected by 3mL of the abdominal aorta after anesthesia.The myocardial tissue below the ligation line was quickly collected,some stored in 4%polyformaldehyde for HE staining,the other part adopted Western blot to detect the expression level of Bcl2,Bax and Beclinl.Results:Compared with the model group,moxibustion group 4,5,7 days heart infarction area percentage decreased significantly(P<0.05),4 d was the best.Compared with the sham operation group,model group's fracture,myocardial fibers,and inflammatory cell infiltration;the level of cTnT,Bax/Bcl2,Beclinl increased at ischemia and reperfusion period(P<0.01).Compared with the model group,at moxibustion group 4 d and autophagy inhibitor group,myocardial fibers arranged in disorder to a lesser extent and with a small amount of interstitial edema and infiltration of inflammatory cells;the level of cTnT,Bax/Bcl2,Beclinl at ischemia and reperfusion stage significantly decreased(P<0.01).Compared with the autophagy inhibitor group,the 4 d moxibustion group's cTnT,Bax/Bcl2,Beclin1 were increased(P<0.05).At each group,there was no significant difference in cTnT,Bax/Bcl2 and Beclinl between the reperfusion and the ischemic stage.Conclusion:Moxibustion with Seed-Size Moxa Cone at Neiguan can effectively reduce the area of myocardial infarction,which is better at 4 days and the protective effect is related to autophagy,which may promote autophagy during ischemia and inhibit autophagy during reperfusion to alleviate the MIRI.
