To investigate the senescence is always a focus in geriatric medical and biological studies,and to reveal the mechanism of senescence can delay the aging speed and extend the life fundamentally.Klotho was identified as an anti-aging gene in 1997 and has inspired tremendous interest in the search for life extension.Klotho gene knockout mice display extensive human aging phenotypes,whereas the transgenic mice over-expressing klotho have approximately 20%extension of their lifespan.Therefore,studying the regulation of klotho gene expression by drugs is very significant for revealing the anti-aging mechanism of drugs.Resveratrol is a natural polyphenol found in a variety of plants.Pharmacology research had demonstrated that resveratrol offered various benefits for common age-related diseases,such as obesity,diabetes,cancer,and neurodegenerative and cardiovascular diseases.However,the anti-aging activity of resveratrol and its effects on the expression of klotho gene in kidney and brain have not been investigated so far.In this study,the anti-aging activity of Resveratrol and its regulation of klotho gene expression were evaluated systemically in vitro and vivo.1.The antioxidant activity of resveratrol was investigated by DPPH,ABTS and FRAP methods.The results indicated that resvertrol showed promising antioxicant activity was detected by DPPH,ABTS and FRAP,which was comparable with that of the well-known antioxidant,ascorbic acid.2.The anti-aging activity of resveratrol was measured the contents of superoxide dismutase?SOD?,catalase?CAT?and malondialdehyde?MDA?in TCMK-1 cell by ELISA method.The results showed that resveratrol could significantly inhibit the formation of MDA and raise the activities of SOD and CAT in TCMK-1.Moreover,resveratrol could repair the TCMK-1 cell damaged by H2O2.3.The anti-aging capability was measured in D-gal-induced aging mice model,and the experimental data showed that resveratrol could significantly inhibit the formation of MDA and raise the activities of SOD and CAT in mice,which was consistent with these of results found in TCMK-1 cell.The RT-PCR,q-PCR showed that the resveratrol significantly increase the expression of anti-aging gene klotho in mice kidney and brain.Histopathological experiment shows that resveratrol could effectively improve the renal and cerebral lesions induced by D-galactose.The results showed that resveratrol exerts anti-aging activity mainly through three ways:?1?removing the free radicals in the body,?2?strengthening the ability of the body by mediating antioxidant enzyme activity and avoiding oxidative stress damage,?3?increasing significantly the expression of anti-aging gene klotho in mice kidney and brain.
