Aging is a programmed biological process determined by both genetic factors and internal or external environmental factors.On the one hand,modern medical research has found that cardiovascular diseases,cancer,diabetes and other chronic diseases are closely related to the aging.On the other hand,China is the most populous country in the world with a large elderly population.Statistics analysis showed that the population aged 60~+are about 17.3%of the total population by the end of 2017.Moreover,it is estimated that the elderly population of 60~+will reach to482 million by 2053.Therefore,the elderly population has brought heavy burden and tremendous pressure to the whole society and their families.Resveratrol,a natural plant polyphenol,widely exists in grapes,polygonum cuspidate and other plants.A large number of studies have found that resveratrol has many biological activities,such as antioxidant,anti-aging,and has a wide range of applications in anti-aging research.In this study,the subacute aging model of mice was established by injection of D-gal.The protective effects of resveratrol against D-gal-induced aging mice were evaluated comprehensively in three aspects including Anti-aging,brain protection and cardioprotection,and the mechanisms involved.The main conclusions of this study are summarized as follows:1.Resveratrol has no effect on the body weight of D-gal-induced aging mice in this experimental time with no significant difference among groups of mice.Compared to D-gal group,resveratrol could improve organ index of liver,spleen and thymus,and promote organ health of aging mice.Resveratrol could increase the level of antioxidant enzymes in the liver of aging mice,increased the activity of T-SOD,GSH-Px and CAT,reduce the content of MDA,and produced antioxidant effect on aging mice.It could also reduce the content of glutamic oxaloacetic transaminase and glutamic-pyruvic transaminase,and promoted the health of liver function.It could also promote the normalization of serum biochemical indicators.The results showed that resveratrol could improve liver health,serum biochemical indexes and immune function of aging mice.In short,resveratrol protected the aging mice.2.Resveratrol increased the brain organ index of D-gal-induced aging mice.Resveratrol could effectively inhibit the apoptosis of brain neurons in aging mice,and significantly reduce the apoptotic rate;significantly reduce the ROS level of brain neurons;resveratrol could protect brain neurons through mitochondrial mechanism,it could increase the mitochondrial membrane potential and the expression of mitochondrial cytochrome C in brain neurons,inhibit the expression of cytochrome C in plasma,and significantly reduce Caspase-9 and Caspase-3 activities.These results showed that resveratrol could protect brain neurons,promote brain health and protect the brain of aging mice through mitochondrial mechanism.3.RSV could improve the index of cardiac organs,the pathological structure of heart tissue,the integrity of cardiac structure,the orderliness of cardiac fibers and the plumpness of cardiac myocytes in aging mice.RSV could significantly reduce the apoptotic rate of myocardial cells,reduce the ROS content and increase MMP of myocardial cells.In addition it increased the T-SOD,GSH,CAT activity and MDA content of heart,improved the antioxidant capacity of heart.Further studies showed that RSV could reduce the levels of TNF-alpha and IL-1beta in myocardial cells of aging mice and inhibit the expression of TLR4,NF-kappa B p65 and Notch 1.That is to say,RSV with protective effect on heart injury in aging mice,and its mechanism is closely related to Notch signaling pathway.
