| Objective:Duplicate infertility mouse model,observe how yi shen tong luo recipe affect mouse testis SOD,MDA level and sex hormoneFSH?LH?T,discuss about the infertilty theraputic machanism of yishen tong luo recipe.Methods:60 male SD rats were randomly divided into 2 groups according to the random number table,the normal group(n = 15)and model group(n = 45).The normal group was given 0.9% normal saline,and the model group was replicated with 500mg/kg/d intragastric administration of purine 15 d.At the end of the model,the two groups were randomly selected to obtain the results of anesthesia,biopsy and pathological examination in order to verify whether the model was successful or not by using.The remaining rats were normal group,the model group was randomly divided into 4 groups,namely,model group of 9,Yishen Tongluo Fang high,medium and low group of each of the 9.Yishen Tongluo group were treated with 20.92g/kg/d,10.46g/kg/d and 5.23g/kg/d respectively for continuous intragastric administration of 30 D.The general state of the rats was observed and recorded daily.12 hours after the first dosage,anesthesia the rats through intraperitoneal injection with 4% chloral hydrate(0.8ml/100g),make the pathological section of the right side testicular tissue of the rats,observe the morphological histology change of the testises.prepare the left sides testis tissue homogenate sample,and test the SOD activiness anf MDS quantity of it.Draw aorta abdominalis blood,centrifuge the sample,utikize serum for sex hormone.Results: 1.Normal group testis pathology(inage 8): rats seminiferous tubule array closely,the borderline can be defined clearly,the construction is completed,mature sperms are developing actively in the tubule,no pathological change in interstitial blood vessel.Pathological models comparion of testis(image 7): the number of testicular spermatogenic cell layers reduce the cell layer is thinner and array disorderly,part of testicular spermatogenic cell core are changed,shrinked,only remained support cells can be found,the number of mature cell in the tubule is reducing.Edema and change can be find in part of the interstitial cell.Yishen tong luo recipe model group(image 4-6)testis pathological sign: the rats seminiferous tubule arrays properly,wall layer construction is decent,spermatogenic cell increased,mature sperm can be find in the tubule.interstitial cell dispersive distribution and number is normal,and more compared with model group in quantity aspect,specially,yishen tong luo recipe model improve more obviously.2.The MDA quantity of rats tistis model is more than normal model(p<0.01),observably,the SOD quantity is lower than normal group(p<0.01).MDA quantity of the medium and high dosage yishen tong luo recipe group is lower than the model group,SOD quantoty is signigicantly higher model group(p<0.01),only slight difference SOD,MDA quantity can be find in low dosage group,which is on statistical significance.3.Compared with normal group,the serum T.FSH level of model comparison group and medium herbal dosage group reduced obviously,the level of high dosage group is lower than normal group,no significant difference betweet two geoups.Compared with model group,significant serum T,FSH,LH average level difference can be found in high herbal dosage group,slight difference in low dodage group.level is dufferent with model group,(p<0.01).Compared with high dosage group,low dosage group FSH,LH level is different averagly(p<0.01),no significantly difference between medium dosagegroup and high dosage group.Conclusion:Yishen tong luo recipe can fix the oxidatibe stress injury that caused by adenine,recovery the SOD,MDA level of rats,balance the change of sex hormone T,LH,FSH level caused by adenine,hence improve the inner environment of rats reproduction,high dosage group perform better than low dosage group. |
