The Effects And Mechanisms Of The Liang Run Tong Luo Recipe On Gut Hormone Of Diabetes Mellitus Gastroparesis Model Rats

Objective: Diabetes mellitus gastroparesis (DGP) is one of common complication of diabetes mellitus (DM). DGP affects the kinetics of blood glucose reducing drugs, is also affects the curative affect its curative effects, leading to blood glucose unstable. For the present, the western medicines adopt gastrointestinal power drugs to treat the disease. It has remarkable curative effects recently, but the disease recurs without taking the drugs and the drug's by-effect is heavier. According to Chinese medicine's traditional understanding to DGP, we formulated the recipe of"Liang Run Tong Luo". To make definite curative effect and mechanism of the recipe in improving DGP, we manufactured DM pattern with STZ injection. With the use of radioimmunoassay and immune group dividing, we examined the MOT, CCK, SS of the blood and the tissue, to probe into its influence of the gastrointestinal hormone and gastrointestinal endocrine cell.Methods: 68 male Wistar rats were divided into four groups randomly: normal control group, model control group, Cisapride group, and LiangRunTongLuo group. The model was established by abdominal administration with STZ. After the establishment of DM animal model, there were not any therapy were adopted. By the end of 12th week, two rats of each group were taken suction to be taken examination. After 12 weeks feeding by common bait vessel, all of the rats were given intragastric administration with different drug by 6 weeks. Stopping given drugs for 24 hours, the blood were collected from femoral artery, and then MOT,CCK,SS were tested with radioimmunoassay. At the same time, the tissue samples, including gastric antrums, duodenum were collected, incretory cells of MOT , CCK , SS were inspected by immunohistochemical method.Result: As fellows.1 The blood glucose of each group of DM rats were higher than the blank group (P<0.01). And there was no difference between the two treatment group (P>0.05).2 Compared with the blank group, the weight of other group significantly decreased (P<0.01). By the 18 week, the weight of the two treatment group increased than the model group. But there was no difference between the two treatment group (P>0.05).3 Compared with the blank group, the food intake of each DM groups were significantly higher after one week (P<0.01). But the reduction was significantly by the 6week, 12week and 18 week (P<0.01). There was noticeable distinction between the two treatment group by the 18 week (P<0.01).4 The comparison of MOT The content of MOT in plasma of each DM group were significantly higher than that of the blank group. But compared with the model group, the content of MOT in plasma of the two treatment group was marked lower (P<0.01). And the content of MOT of LiangRunTongLuo group was even better than that of the Cisapride group (P<0.05).5 The comparison of CCK The content of CCK in plasma of each DM group were significantly higher than that of the blank group. But compared with the model group, the content of CCK in plasma of the two treatment group was marked lower (P<0.01). And the content of CCK of LiangRunTongLuo group was even lower than that of the Cisapride group (P<0.05).6 The comparison of SS Compared with the blank group, there were noticeable decrease in the content of SS in plasma of each DGP group. But compared with the model group, that of the two treatment group significantly increased (P<0.01). And that of the LiangRunTongLuo group was even higher than that of the Cisapride group (P<0.01).7 The immunohistochemical staining result of MOT in small intestine of rat:⑴Morphological observation: There were some dilute brow reactant in endochylema of immunological positive Mo cells, which local in the epithelial of duodenal glands and bottom of villi. And the cross-section of the cells were seen as rounded, oval and shuttle, etc.⑵Quantity of Mo cells in optical microscope: Compared with the blank group, the quantity of Mo cell of the model group were significantly lower, but the hadro-positive cell rate were noticeable higher (P<0.01). There was no difference between the two treatment group with the blank group (P>0.05). The quantity of Mo cell of LiangRunTongLuo group and Cisapride group were higher than that of the model group(P<0.01), but the hadro-positive cell rate of the two treatment group were much lower than that of the model group(P<0.01). And there was no difference between the two treatment group in both quantity and rate of Mo cells (P>0.05).8 The immunohistochemical staining result of CCK in small intestine of rat:⑴Morphological observation: There were some dilute brow reactant in endochylema of immunological positive CCK cells, which local in the epithelial of duodenal glands and bottom of villi. And the cross-section of the cells were seen as rounded, oval and shuttle, etc.⑵Quantity of I cells in optical microscope: Compared with the blank group, the quantity of I cell of all the DGP group were significantly higher. the hadro-positive cell rate of the model group and the Cisapride group were noticeable higher (P<0.01). But there was no noticeable difference with the LiangRunTongLuo group (P>0.05). Compared with the model group ,the quantity of I cell and the hadro-positive cell rate of LiangRunTongLuo group was much lower (P<0.01). But there was no difference between Cisapride group with the model group (P>0.05). The quantity of I cell and the positive rate of LiangRunTongLuo group were much lower than that of Cisapride group (P<0.01). 9 The immunohistochemical staining result of SS in gastric antrum of rat:⑴Morphological observation: There were some dilute brow reactant in endochylema of immunological positive D cells, which mainly local in the middle or underlayer of gastric antrum of rat. And the morphous of D cell was irregular.⑵Quantity of D cells in optical microscope: Compared with the blank group, the quantity of D cell of all the DPG group were significantly higher, but the hadro-positive cell rate were noticeable lower (P<0.01). The quantity of D cell of LiangRunTongLuo group was much lower than that of the model group, and the positive rate was much higher (P<0.01). There was no difference between Cisapride group with the middle group in that two directions (P>0.05). The quantity of D cell of LiangRunTongLuo group were significantly lower than that of the Cisapride group ( P < 0.01), but the hadro-positive cell rate was much higher than that of the Cisapride group (P<0.01).Conclusions: 1 The method with STZ injection and long-time feeding on rats which duplicate the animal model is successful, and the model is steady, simple and economical. 2 According to main pathogenesis of diabetes mellitus gastroparesis, we instituted the Liang Run Tong Luo recipe, which was proved by experiment that it has the superiority in improving the obstacle of the gastrointestinal function and adjusting of Gut Hormone. 3 By the time of eighteenth week, the rats presented the disorder of MOT,CCK,SS. Liang Run Tong Luo recipe, as compound preparation, affected the excretion and releasing of sorts of gut hormone, the target range is wider, which superior to Cisapride. 4 Adjusting stomach intestine function to influence gut hormone, may be one of the mechanism of the recipe on improving the obstacle of gastrointestinal function.
