Objective: This topic through the construction of skin and soft tissue defect wound in rabbit ears model,using self-made miniature vacuum sealing drainage device for rabbit ear wound to make the experimental study of angiogenesis,to verify the Mini VSD on wound treatment effect,to explore its validity and feasibility in clinical medicine,and provide theoretical and scientific experimental basis for clinical application of Mini VSD.Methods: Take 20 rabbits,and weighted 2.0~3.0kg,regardless of male and female.On the day of the experiment,the knife removed ears back hair,10% chloral hydrate(4ml/kg)intraperitoneal injection of anesthesia,fixed in prone position.With 0.9% saline cleaning rabbit bilateral ears,and then 75% ethanol disinfected them.The circular area marked with diameter about 2cm between central neurovascular bundle and ear vein of rabbit ear,and resected of full-thickness skin,retained only the cartilage membrane,with a cotton ball compressing hemostasia.The left ear wound was treated as the treatment group,and the right ear wound was the control group.Cut the speed of the micro dynamic dressing into 2cmx2 cm size and placed it in the wound site.Took 2 infusion tubes with a diameter of about 2mm,one of them was used as drainage,and 3 side holes were cut at the end of the tube,and the other was used to flush the wound.At the same time,the wound and the dressing was completely sealed with transparent film,drainage tube by a margin of skin edge led to connect central negative pressure or 50 ml syringe,every day to 10 kp pressure to be continuously drainaged every day,continuous suction for 8 hours.Another irrigation tube,connected to the 20 ml syringe,was humidified 3 times a day.Control group wound: covered with ordinary Vaseline gauze and dressing.After treatment,the animals were placed in a rabbit cage to avoid interference with the wound surface.Postoperation of wounds 1,3,5,7,9 days took pictures after treatment and wound dressing changed,and calculated the rate of wound granulation tissue in wound healing.And the samples were stained with HE,to observe the pathological changes of histology,and immunohistochemistry of VEGF,measured the number of newborn rabbit ears wound at different time points in the blood vessels,finally the results were made for statistical analysis.Results:1.The rate of wound healing at different time points after operation: Compared with the control group,the treatment group promoted wound healing in rabbit ear rate was significantly better than the control group(P<0.05);and with the extension of time,the treatment group and the control group the healing rate of increasingly obvious differences,to ninth days,the control group wound healing rate was 60.8 + 2.7%,the treatment group arrived at the healing state and wound healing rate of 81.9 + 5.2% in the treatment group,the difference was statistically significant(P<0.01).2.Granulation tissue of wound surface at different time points after operation: The third day,5th day,7th day,the gap between the treatment group and the control group of granulation tissue content was increased,the control group of granulation tissue was significantly lower than the treatment group,and ninth day,the content of granulation tissue treatment group is 4.13 + 0.48,while the control group reached 2.91 + with granulation tissue 0.37,t h e d i f f e r e n c e w a s s t a t i s t i c a l l y s i g n i f i c a n t(P < 0.0 1).3.Wound neovascularization density: Microscopically,the treatment group first day,a margin of shallow tissue showed a small positive expression of VEGF in vascular endothelial cells and gradually with the prolongation of treatment time,the positive expression of VEGF increased significantly in dermal vascular endothelial cell wound tissue,the wall was brown,lumen was round,visible red blood cells in the lumen.A small amount of perivascular infiltration of inflammatory cells.Fifth days of treatment of vascular density increased significantly,seventh days of wound vessel density reached the highest value,to ninth days of vascular density gradually stabilized,vascular density in treatment group was significantly higher than the control group,the difference was statistically significant(P<0.01)..Conclusion: The application of miniature vacuum sealing drainage device is feasible and effective.It can promote endothelial cell migration,proliferation,angiogenesis,promote the growth of granulation tissue and wound healing.It has curative effect.
