Objective Through the determination of protein expression level and mRNA expression level of Matrix Metalloproteinase-9(MMP-9)and its specific inhibition--Tissue Inhibitor of Metalloprotease-1(TIMP-1)of lung tissue in rats with chronic obstructive pulmonary disease(COPD),to further study effect and action mechanism of polygonum cuspidatum.Methods A total of 50 SPF healthy male SD rats were divided randomly into norma 1 control group(10 cases),COPD control group(10 cases),prednisone intervention grou p(10 cases),polygonum cuspidatum intervention group(10 cases)and prednisone comb ined with polygonum cuspidatum intervention group(10 cases).Smoked and endotrache al lipopolysaccharide injection is adopted to establish the rat COPD model,and the s moke of 29 days using drug related intervention,Closely observe and record the rats in the case of the experiment,to observe weight,after the experiment,snapping the ra ts executed sampling,collecting specimens of lung tissue,observe each rat lung tissue pathology change,Groups of rats was evaluated by western blot method MMP-9 and TIMP-1 protein expression,the application of real-time fluorescence quantitative PCR m ethod between groups rats MMP-9 and TIMP-1 mRNA expression level.All the data using SAS9.4 statistical software for statistical processing.Results 1.The weight change before and after the experiment:Before the experiment,weight of 5 groups rats has no significant differences(P>0.05);After the experiment,COPD rats weight of four groups was significantly lower thanthat of normal control group(P<0.05);After the experiment,after drug intervention,weight of polygonum cu spidatum intervention group and prednisone combined weight knotweed intervention gr oup was significantly higher than that of COPD control group(P<0.05),weight of pol ygonum cuspidatum intervention group increased significantly(P<0.05),and weight had no significant difference between prednisone intervention group and COPD control gro up(P>0.05).2.pathologic change:After buliding,the lung tissue pathology showed tha t accord with COPD pathologic change,its after drug intervention from COPD control group to reduce the lung tissue pathological changes,including prednisone combined polygonum cuspidatum intervention group works best.3.MMP-9 and TIMP-1 protein e xpression of lung tissue:Compared with normal control group,MMP-9 and TIMP-1 protein expression levels of COPD rats of four groups were significantly increased(P<0.05);Compared with COPD control group,MMP-9 and TIMP-1 protein expr ession levels of prednisone intervention group,polygonum cuspidatum intervention gro up and prednisone combined polygonum cuspidatum intervention group were significant ly lower(P<0.05),of which the prednisone combined polygonum cuspidatum group M MP-9 and TIMP-1 protein expression to reduce the most significant(P<0.05).4.MMP-9 and TIMP-1 mRNA expression of lung tissue:Compared with normal control group,MMP-9 and TIMP-1 mRNA expression levels of COPD rats of four groups were significantly increased(P<0.05);Compared with COPD control group,MMP-9 and TIMP-1 mRNA expression levels of prednisone intervention group,polygonum cuspi datum intervention group and prednisone combined polygonum cuspidatum intervention group were significantly lower(P<0.05),of which the prednisone combined polygonu m cuspidatum group MMP-9 and TIMP-1 mRNA expression to reduce the most signif icant(P<0.05).Conclusions ? traditional Chinese medicine(TCM)polygonum cuspidatum effective intervention model rat COPD airway remodeling,and to some extent can relieve clinic al symptom,its mechanism of action is to:polygonum cuspidatum can from protein a nd gene level to correct the COPD rats protease and resistance to protease imbalances;? the prednisone combined with polygonum cuspidatum more effective than predniso ne alone. |
