Purpose:Impacts on the Contents of IgA1?TfR?TGF-?and IL-6 of IgA Nephropathy Model RatsTreated with the Therapy for Tonifying the kidney Bushenjianpi and Jieduliyan,to study Bushenjianpi and Jieduliyan method on treating IgA nephropathy rat the influence of kidney tissue morphology and immune deposits,to study the efficacy of the treatment of IgA nephropathy mechanism.Method:A total of 80 healthy male Wistar rats of clean grade,weight?180±20?g,Adequate feeding after feeding for 1 week,the rats were divided into blank group?n=10?and model group?n=70?,blank group:from the beginning of the experiment,with 4ml/kg of distilled water every other day,lasted for 8 weeks.Subcutaneous injection of 0.9%sodium chloride solution 0.4ml/every one,once a week for 9 weeks.At the end of the 6th week,intravenous injection of 0.9%sodium chloride solution 0.2ml/every oneFrom day 10,give4ml/?kg·d?saline,once a day for 12 weeks.The model adopts Lu Huiyu[1]reported methods,Bovine serum albumin?BSA?crystal powder was mixed with distilled water at a concentration of 100 g/L,and the rats were incubated at 4 ml/kg every other day for 8weeks from the beginning of the experiment,Carbon tetrachloride?CCL4?and castor oil were mixed at a ratio of 1:3 to 0.4 ml/subcutaneously once a week for 9 weeks,Lipopolysaccharide?LPS?was mixed with 0.9%NaCl in 0.25 g/L solution and 0.2 ml was injected into each tail vein at week 6.At the end of the 9th week,12 rats were randomly selected,including 10 in the model group and 2 in the blank group.The observation index:?1?To test the 24-hour urine red blood cell count and 24-hour urine proteinuria quantitative changes;?2?Detection of serum renal function and liver function in rats;?3?Were measured with ELISA method for the determination of serum and renal tissue IgA1,TfR,TGF-?,IL-6;?4?Using immunofluorescence to detect IgA complex deposition in the kidney tissue.Result:?1?During the experiment rats were killed 22,including 12 killed validate the model,modeling died during the 10.?2?The rats in the model group were treated with subcutaneous injection of carbon tetrachloride,and the rats in the model group showed lethargic anddiarrhea after subcutaneous injection.At the same time,Rat nose and mouth,superficial skin at the Department of bloody secretions,rat weight also showed a significant decline in performance.After 2 to 3 weeks of treatment,the rats in the treatment group were improved in general condition,diarrhea symptoms decreased and weight gain.?3?The rats in the blank group had no hematuria and proteinuria symptoms.The rats in the model group had a significant increase in RBC and 24 hours UTP in the model group?P<0.05?.The treatment group And Luoding new control group were significantly different?P<0.05?.?4?Blank group:In the rat kidney tissues,in addition to the vascular endothelium,did not see the deposit of IgA molecules.Model groups:Can be obviously of rat renal tissues,IgA molecules formed by the combination of linear and fluorescent material or punctiform fluorescence,the fluorescence material into linear array.?5?IgA nephropathy model rat renal tissues,visible obvious IgA1,TfR,TGF-beta,IL-6 molecular deposition compared with blank group increased significantly.?6?Each treatment group rats after treatment of IgA1,TfR,TGF-?,IL-6 levels significantly lowered?P<0.05?,the comparison between groups,kidney and spleen,detoxification,and los Ding Xin pharynx high,low and middle dose group.The control group compared with significant difference?P<0.05?.?7?There was significant difference between groups of renal function and liver function in rats?P>0.05?.Conclusion:?1?Methods using bovine serum albumin gavage and carbon tetrachloride,LPS injection could successfully establish animal model of IgA nephropathy.?2?Bushenjianpi and Jieduliyan group effectively reduce IgA nephropathy model rats24hours urine red blood cell count;Lotensin group and Bushenjianpi and Jieduliyan group effectively reduce IgA nephropathy model rats 24 hours urinary protein excretion rate.?3?The levels of IgA1,TfR,TGF-?and IL-6 in renal tissue of IgA nephropathy rats were significantly higher than those in blank group,and the levels of IgA1,TfR,TGF-?and IL-6 were decreased after administration?P<0.05?.Conclusion The abnormal expression of IgA1,TfR,TGF-?and IL-6 is involved in the pathogenesis of IgA nephropathy,leading to the occurrence of IgA nephropathy.?4?Each treatment group,Bushenjianpi and Jieduliyan prescription possible mechanism by adjusting the level of Th type cytokines IgA1?TfR?TGF-??IL-6 in renal tissue of rats IgAN model,to delay the development of Ig AN.
