Purpose Discuss the impact of TSH on the reproductive endocrine disorder when one is with hypothyroidism by studying the effect of TSH on estradiol seceretion function and the receptors on membrane of COV434 cells.Method Culture cells for 48 h by treated with 0.01?0.1?1U/L of TSH and constant concentration of FSH,cells receiving no TSH stimulation were taken as control,detect the estradiol level of cells supernatant fluid by enzyme-linked immuno sorbent assay,detect the expression of the FSH receptor and TSH recrptor mRNA by polymerase chain reaction,detect the protein expression level of the FSH receptor on membrane by western blot.Result Compared with control group,when the concentration of TSH is above 0.1 U/L,the estradiol level decreases visibly(P<0.05)and the expression level of TSH receptor mRNA increases visibly(P<0.05);The expression level of FSH receptor mRNA and protein has no significant change with control group(P>0.05).Conclusion When the concentration of TSH is above a certain level,TSH has a negative impact on the endocrine function of COV434 cells;The phenomenon could be related to the change of expression level of TSH receptor.FSH receptor has no concern with the change of concerntration of TSH,no matter from the level of protein or gene.
