Objective:To investigate the effects of apolipoprotein M(ApoM)on the expression of NF-κB,interleukin-1β(IL-1β)and intercellular adhesion molecule-1(ICAM-1)and interleukin-6(IL-6)mRNA levels in myocardium of mice.Methods:Using base quenched probe techniques,we confirmed the genetypes of mice as ApoM wild type(ApoM+/+)or ApoM knockout type(ApoM-/-).ApoM-/-mice were randomly divided into control group and other 2 groups(1 h,3 hrs.)with 6 mice in each group.ApoM+/+mice were also grouped in the same way.The mice of control group were not accepted any treatment.The mice in other 2 groups were randomly assigned to receive intraperitoneal injection of lipopolysaccharides(LPS)(2mg/ml)and mice were sacrificed at 1 h,3 hrs respectively.The myocardium tissues were collected and detected mRNA expression of NF-κB,IL-1β,IL-6 and ICAM-1 by Polymerase Chain Reaction.Eighteen ApoM-/-male mice aged 6 to 8-weeks-old(approximately 2325g body weight)and eighteen age and gender matched ApoM+/+mice were applied in the present study.Results:With the effect of LPS,the levels of NF-κB,IL-1β,IL-6 and ICAM-1 in ApoM-/-mice myocardium were significantly higher than ApoM+/+mice myocardium,1h(all U values are 0.00;all P values are 0.00);3h(U value:3.00,0.00,0.00,1.00;P value:0.015,0.002,0.002,0.004).TWO-Way ANOVA showed that there was interaction between LPS and ApoM on NF-κB,IL-1β,IL-6 and ICAM-1 expression levels in mice myocardium.1h(F value:23.5,33.74,40.90,167.00;P value:<0.001,<0.0001,<0.0001,<0.0001);3h(F value:6.092,27.91,12.66 and 12.06;P value:0.023,<0.0001,0.002,0.002).Conclusions:ApoM inhibited LPS-induced NF-κB,IL-1β,IL-6 and ICAM-1expression in mice myocardium. |
