Objective: To detect the expression of P16 and SOX-2 in cervical intraepithelial neoplasia(CIN)by immunohistochemistry and to explore the difference of expression of P16 and SOX-2 in CIN.CIN diagnosis and treatment to provide a certain theoretical basis. Methods: Eighty pathological specimens of cervical intraepithelial neoplasia diagnosed by pathology department of Duxian People's Hospital from June 2013 to June 2016 were selected and 40 cases were diagnosed by pathology department of Du County People's Hospital For normal tissue samples of cervical tissue.According to the different degree of cervical intraepithelial neoplasia, the low grade cervical intraepithelial neoplasia(n = 30),the high grade cervical intraepithelial neoplasia(n = 50).At the same time, normal cervical tissue samples were set as normal group(n = 40).Immunohistochemistry was used to detect the expression of P16 and SOX-2 in cervical intraepithelial neoplasia and normal tissues. All data in this study were analyzed by SPSS 20.0 statistical software, and Spearman test was used to analyze the correlation.Results:(1)The positive rates of SOX-2 and P16 in normal tissues were all 0.00%,while the positive rates of SOX-2 and P16 in cervical intraepithelial neoplasia were 55%(44/80)and 67.5%(54/ 80).The positive expression rate of SOX-2 in low-grade group was 13.33%(4/30)and the positive expression rate of P16 was 23.33%(7/30).The positive expression rate of SOX-2 80%(40/50),the positive expression rate of P16 was 94%(47/50).The positive rates of P16 and SOX-2 in the two groups were significantly different,P <0.05,the difference was statistically significant.(2)The positive expression of P16 was found in 44 SOX-2 positive cervical intraepithelial neoplasia specimens, and the positive expression rate of SOX-2 and P16 was significantly correlated(P <0.05).Conclusions:(1)P16 and SOX-2 can be used as biomarkers for differentiating normal cervical tissue from cervical intraepithelial neoplasia. The positive expression rate of P16 and SOX-2 with the increase of the degree of cervical intraepithelial neoplasia Rise.(2)The positive expression rate of P16 and SOX-2 in cervical intraepithelial neoplasia has obvious correlation. The combined detection can objectively reflect the degree of cervical intraepithelial neoplasia and improve the accuracy of pathological diagnosis.
