Sarcandra Glabra Sanqing Granule,a Chinese veterinary compound preparation,was developed by Guangxi University and Guangxi Beidouxing Animal Health Pharmaceutical Co.,Ltd.The purpose of this study was to investigate the in vitro anti-inflammatory effect,protective effect on asthmatic mice and the effect of reducing urine to further clarify its pharmacodynamic effects of Sarcandra Glabra Sanqing Granule,and also to investigate its reliability via mice acute toxicity test and rats subchronic toxicity test.Methods:(1)Effect of Sarcandra Glabra Sanqing Granule,in a certain range of concentration,on cell viability and LPS-induced TNF-alpha and IL-6 secretion were performed through MTT assay and ELISA assay in RAW264.7.(2)A murine model of asthma was established,and then detected different concentrations of Sarcandra Glabra Sanqing Granule on IL-4,IL-5,IL-13 secretion and inflammatory cells production in BALF;Likewise,specific IgE level in serum of mice and mice airway hyperresponsiveness were then measured.(3)Mice hyperuricemia model was also established,effect of Sarcandra Glabra Sanqing Granule and its individual components on serum uric acid and liver xanthine oxidase activity in mice were performed.(4)Multiple gavage was performed during 24 hours to evaluate half lethal dose(LD50)of Sarcandra Glabra Sanqing Granule in mice.(5)Finally,the rats were subjected to intragastric administration for a period of 35 days;The changes of body weight,organ coefficient,blood routine,serum biochemical indexes were assayed and histopathological changes were observed.Results:(1)compared with the control group,cell viability of RAW264.7 in each groups at different concentrations of Sarcandra Glabra Sanqing Granule were significantly increased,and LPS induced pro-inflammatory cytokines TNF-alpha,IL-6 secretion in RAW264.7 cell were inhibited.(2)Compared with the control group,three different concentrations of Sarcandra Glabra Sanqing Granule could reduce the number of inflammatory cells in mice BALF and significantly inhibit secretion of IL-4,IL-5,IL-13 and serum specific IgE level,thus reduce airway hyperresponsiveness in mice.(3)compared with the positive control group,Sarcandra Glabra Sanqing Granule and Sarcandra Glabra Granule could significantly decrease the serum uric acid level and liver xanthine oxidase activity in mice.(4)acute toxicity test failed to detect LD50 of Sarcandra Glabra Granule on mice;At the maximum dose of 2.4 mL/20g(equivalent to 75g per kg body weight)mice did not exhibit any toxic response.(5)in the subchronic toxicity test of rats,body weight,organ coefficient,blood routine and blood biochemical index of rats in groups at different concentrations of Sarcandra Glabra Sanqing Granule had no significant difference and no abnormal pathological were changed when compared with the control group.Conclusion:Sarcandra Glabra Sanqing Granule exhibit predominant in vitro anti-inflammatory effect and satisfactory protective effect on asthmatic mice.It could significantly reduce the serum uric acid and liver xanthine oxidase activity in mice hyperuricemia model and show no acute toxicity and subchronic toxicity to mice.Our study suggest that Sarcandra Glabra Sanqing Granule is safe and effective Chinese herbal medicine preparation.
