Objectives:Traumatic optic nerve injury is common in clinic,which causes obvious symptoms such as Visual impairment and visual field defect.However,the prognosis of clinical treatment is not clearIn order to formulate efficacious therapies,it is essential to figure out the pathogenesis of various types of traumatic optic nerve injury.Many researchers have previously conducted relevant studies of animal models,which were made by hitting the animals' head,clamping the optic nerve,pulling the optic nerve,and squeezing the optic nerve.The degree of damage,the time of damage,and the damage site were uncontrollableBased on above,the fundamental objectives of this study is to show solicitude for the following two aspects:firstly,three new types animal models of traumatic optic nerve injury with different degrees of injury are successfully established;then analysing the functional,morphological,and molecular cytological difference of these animal modelsMethods:(1)Three different models of traumatic optic nerve injury were made by injecting sodium hyaluronate beneath optic nerve sheath,clamping the optic nerve with neoteric clip and inserting bone fragment into the optic nerve.(2)Utilizing the visual evoked potential(P-VEP,F-VEP)to monitor the functional changes of these three different models.(3)Pathological section and HE staining were used to study morphological changes of three traumatic optic nerve injury models.(4)immunohistochemistry(Caspase-3/P53/Bax),horseradish peroxidase labeling and TDT mediated dUTP nick end labeling(TUNEL)were employed to discuss molecular cytological difference of these animal modelsResults:(1)Successfully produced three diverse animal models of traumatic optic nerve injury.(2)Compared with the normal group,visual evoked potential was notablly changed and pathological section and HE staining revealed different degrees of damage among three models.Outcomes of immunohistochemistry were markedly rised,apoptotic cell increased and axonal transport was obstructedConclusion:Three new animal models of traumatic optic nerve injury have been successfully constructed from three aspects:functional,morphological and molecular cytology.
