Purpose: To study the relationship between biofilm formation and antibiotic resistance of Chinese medicine resistant Escherichia coli.Method: 1?The tested UPEC strains(1351,4996,5028)were isolated clinically from a hospital at the Hebei Medical University,and showed various resistance to a Chinese herbal extract.The control strain Trans 10 was the standard strain of Escherichia coli;2?Biofilm measurements were performed with 96-well plate crystal violet staining and OD value detection;3?Preparation and transformation of competent cells by OD value detection,electroporation and resistance gene screening;4?Construction of luxS gene knockout strain by real-time fluorescence quantitative PCR,gene homologous recombination and agarose gel electrophoresis.Result: We found that the expression of pgaA and pgaB genes was very different in 1351 and 5028 strains,and the lack of luxS gene of the 4996 strains will affect the formation of biofilm.Conclusion: 1.The strains with Chinese traditional medicine resistance are related to the ability of biofilm formation.2,the biofilm formation ability of Chinese medicine resistant strains had different response to the presence of glucose.3.The AI-2 encoded by the luxS gene of the resistant strains of Chinese medicine is not the essence of biofilm formation,but may be a better factor to help biofilm formation.
