Objective1.Establish the invitro biofilm models of drug-resistance Escherichia coli.2.To study the effect situation of Scutellaria baicalensis and self-made traditional Chinese Medicine that respectively combined with ceftazidime on the formation of mature biofilm of drug resistance Escherichia coli, which can be provided experimantal basis for clinical drug.Methods1.Automatic bacteria analyzer can identify it for Escherichia coli, K-B disc diffusion method for determination of susceptibility of Escherichia coli.Applying Alcian blue with Congo red staining and Congo red flat characterization of Escherichia coli biofilm, also take PVC as a carrier, L-B broth96-well plate to establish the invitro model of Escherichia coli biofilm.2.When Escherichia coli cultivates ld,2d,3d,5d,it requires to use semi-quantitative of Crystal Violet biofilm. Scanning electron microscope can observe the surface of carrier to get the information about the formation process of Escherichia coli biofilm,in order to determine the time of Maturation of biofilm.3.The application of micro-dilution to test Scutellaria baicalensis、self-made Chinese medicine and ceftazidime to MIC of clinical Escherichia coli and norm strains, according to MIC values than preparation to prepare different concentrations of drugs. After drug for24hours, use checkerboard method to joined which contain mature biofilm Of Escherichia coli in a96-well plate, by counting observation about living bacteria in the biofilm on effect of mature biofilm of Escherichia coli.Results1.In the DH-360under the condition of electric heating constant temperature incubator B broth is training for3days that the strains of drug-resistant Escherichia coli can form mature biofilm;2.MIC values of scutellaria baicalensis and self-made herbs for experimental strains are32768ug/ml, for the standard Escherichia coli strains respectively are8192ug/ml and16384ug/ml. MIC values of ceftazidime for experimental and standard Escherichia coli strains respectively are32ug/ml and4ug/ml. the minimum effective concentrations of scutellaria baicalensis which combines with ceftazidime take effect on formation mature biofilm of Escherichia Coli are16384ug/ml. the minimum effective concentrations of ceftazidime are4ug/ml. self-made traditional Chinese medicine combines with ceftazidime in acting on the formation of mature biofilm of Escherichia Coli, the minimum effective concentrations of self-made traditional Chinese medicine are8192ug/ml, ceftazidime are4ug/ml.Conclusion1.The biofilm of escherichia coli plays an important role in resistance drug mechanism of Escherichia coli.2.Alcian blue with Congo red staining repeats good, sensitivity high, operation easy, besides, it owns the ability of early sieve for bacteria whether produces biofilm, and can be applied in clinical microbial detection.3.In this experiment conditions, Escheri-chia coli can be trained for72hours to form mature of biofilm.4.self-made Chinese herbs can inhibit the growth of Escherichia coli, By means of drug combination with ceftazidime that can enhance the sterilization effect and reduce their dosage.
