With the development of the discipline such as biology,material science,and analytical chenmistry,advanced technology and mechine have being used in the filed of medical diagnose,which promotes the great progress of medical instruments.Thereinto,the point-of care testing(POCT)device with the merits of miniaturization,simplicity of operator,small sample needed and fast detection efficiency is leading it interested by scientists.Herein,we have intergrated microfluidic technology with structural color coded technology and microbeads array technology,and designed a low-cost chip based on coded mirobead array which can be applicated in multiplex detection of biomolecule.The detailed work descriabled as flollows.(1)The design and preparation of chip.We designed a flow-througth chip,PET and PMMA sheets,porous Ni square mesh grid and transparent double-sided tape materials were used for the preparation of the chip,and beads as carriers be loaded in the chip.The low-cost raw materials,simple fabrication procedure and ease-to-use detection process making the chip is very beneficial for large-scale production.Moreover,we characterized and optimized the feature of the chip,leading the chip more suitable to detecte biomolecule.(2)The application of microbead-array chip in protein detection.First,we discussed the suitable cylcle times of sample solution while the chip was used in detection.And compared the detection result of flow-through and flow-over chip,which shown the advantages of the flow-through chip.Then,we detected the proteins in chip based on fluorescence and Raman spectrum as report signal respectively,which demonstrated the practicability of chip application in protein detection.And the detection limit of the bead-array chip system is 122.6 pg/mL of AFP which is improved greatly compared with the traditional bead-based protein detection methods.(3)The fabrication of angle independent structural color coded beads.In order to achieve the angle independent structural color coded silica microspheres,the carbon black nanoparticles,gold nanoparticles and the electrolyte were added into the silica nanoparticles solution to adjust the structural color,and successfully obtained beads with angle independent structural color.
