ABSTRACTObjective: Under the guidence of TCM theory, the author applies Shuyu Granuls to treating the depression model rats, observes the effect of Shuyu granules on the behavior of depression model rats and judges the effectiveness of treatment. The research is conducted by observing the pathological changes of hippocampus in rats and hippocampus of rats of brain-derived neurotrophic factor BDNF and CAMP response element binding protein CREB and B Cell Leukemia 2 Bcl-2 expression, and exploring its mechanism in the treatment of depression, which clarify the target of traditional Chinese medicine in the treatment of depression and elucidate the relationship between the mechanism and neurotrophic factors, and provide experimental data for clinical, trying to provide a new way for the treatment of depression.Methods:1. 72 Wistar rats of clean grade were randomly divided into 6 groups, 12 rats in each group. The six groups were the control group, depression model group, low dose group of Shuyu granules, Shuyu granules medium dose group, high dose group of Shuyu granules and the positive drug control drug group(Fluoxetine). The chronic unpredictable stress stimulus and solitary were combined to produce depression model rats by behavioral indicators such as weight of the rats and sugar consumption, open field test and swimming time, which were as observation index. The depression model rats were treated by Shuyu granules and the effects were recorded during the administration of the behavior evaluation index.2. Transmission electron microscope and optical microscope are employed to observe the ultrastructure of the hippocampus of rats, then record the effect of Shuyu granules on the hippocampal structure of rat depression model.3. Expression was detected by immunohistochemical method in rats hippocampus BDNF, CREB, Bcl-2 protein.4. The expression of BDNF, CREB and Bcl-2 m RNA in hippocampus of rats was detected by Real-Time PCR.Results:1. Shuyu Granuls can improve the general state of depression model rats.2. Shuyu Granuls can improve the pathological changes of hippocampal neuronsin depression model rats.3. Shuyu Granuls has increased the content of BDNF, BDNFm RNA in hippocampusof depression model rats.4 Shuyu Granuls can improve the expression of CREB, CREBm RNA in thehippocampus of depression model rats. 5 Shuyu Granuls has increased thecontent of Bcl-2m RNA, Bcl-2 in hippocampus of depression model rats.Conclusion:Shuyu granules can improve behavior disorder of depression model rats and influence the organizational structure in hippocampus of depression model rats, increase the content of BDNF in brain in hippocampus of depression model rats, and CREB content and expression of CREB m RNA in depression model rats, and improve Bcl-2 content and Bcl-2m RNA expression in hippocampus of depression model rats, which suggests that Shuyu Granuls take effect as an antidepressant through the intervention of neurotrophic factors, signal transduction and anti apoptosis in rat model of depression.
