Purpose: Through Jieduyizhifang role in alzheimer's disease transgenic c.elegans research,to observe the survival rate of nematode paralysis rate and determine the quantity of related gene expression,to Ming to study the effect of Jieduyizhifang on the educational party nematodes nerve damage mechanism.To provide experimental basis for drug entry.Method: Used to water the Jieduyizhifang of educational experiment extract made from producing to spare,experiment chooses transgenic A?1-42CL4176 elegans as the experimental animal model,the synchronization processing(0 mg/L)were randomly divided into model group,drug low dose group(25 mg/L),middle dose group(50 mg/L),high dose group(100 mg/L).Life experiment in each group 50 nematodes,for every 24 hours to count the number of nematodes deaths,according to nematodes charted the deaths,and four experimental groups of nematodes were the average life expectancy,maximum life span is analyzed.The paralytic rate experiment was performed in each group of 30 nematode worms,and observed the number of paralysis and death of nematode worms every two hours,and the data were analyzed based on the number of non-paralysed Numbers of the nematodes.Selected model group and the middle dose group of nematodes,each group of 100,will be in the same period of nematodes transfer in the training after 48 h 16 ?to 25 ?for induction,induced after 48 h,extracting total mRNA of C.elegans,respectively,after synthesis cDNA reverse transcription,finally the real-time fluorescent quantitative PCR to detect TNFAIP1 homolog(F22E5.6,ZC239.12),HSP16.2(Y46H3A.D),HSP16.41(Y46H3A.E)the amount of gene expression changes.Result:1.The average life span of each drug dose group was 30.8%,22.05%,64.70%,and an average of 39.22%.2.Life expectancy was prolonged,but the survival of nematodes did not rise with the increase in drug concentration.3.Life experiments data statistics as a result,the P value is less than 0.05,the experiment has statistically,and dose group(50 mg/L)than the model group data P values < 0.01.4.The study of nerve damage was determined by A?,and compared with the model group,P < 0.05,there was a statistical difference.5.Low dose group(25 mg/L)nematodes paragraphs time not paralysis rate were higher than in model group nematodes paralysis ratio,of which 76 h when two groups nematodes of paralysis rate difference is biggest,low dose group was 63.33%,the model group was 40%.6.Jieduyizhifang has increased TNFAIP1 homolog(F22E5.6,ZC239.12),HSP16.2(Y46H3A.D),HSP16.41(Y46H3A.E)and other gene transcription levels.Conclusion:1.Jieduyizhifang can extend the lifespan of nematode worms and reduce the rate of paralysis,which is not associated with drug concentration.2.The effect of low dose Jieduyizhifang is better than medium dose and high dose Jieduyizhifang on the protection of nematode nerve injury.3.Jieduyizhifang by raising TNFAIP1 homolog(F22E5.6,ZC239.12),HSP16.2(Y46H3A.D),HSP16.41(Y46H3A.E)such as gene transcription,activate the expression of resistance genes,and the progress of the anti-aging and paralysis.
