| Purpose:To observe the effect of Jie Du Tong Luo Fang on the expression of heart tissue transglutaminase(tTG)in the myocardial fibrosis(MF)rat.Method: Totally 36 adult SPF grade SD male rats were randomly divided into the normal control group,the model group,the ACEI group and Jie Du Tong Luo Fang High,medium and low dose groupThe model group,the ACEI group and Jie Du Tong Luo Fang each dose group Give injections of isoprenaline,rats in the Jie Du Tong Luo Fang each dose group were administered with 10,20,30g/kg Jie Du Tong Luo Fang granule suspension per day respectively by gastrogavage.rats in the ACEI group were administered with 0.005g/kg ACEI suspension per day respectively by gastrogavage,Equal volume of saline was given to rats in the normal control group and the model group by gastrogavage.All the medication was performed once daily for 28 successive days.The heart tissue of rats were withdrawal 24 h after the last intragastric administration?The morphology of the heart was observed using HE and Masson trichrome staining.Hydroxyproline(Hydro)level in myocardium was determined by a colorimetric method.The tTG protein expression of heart tissue observed by immunohistochemistry.The tTG mRNA expression of heart tissue was determined by real-time quantitative reverse transcriptase PCR(RT-PCR).Result:(1)Cmopared with normal control group,the Jie Du Tong Luo Fang each dose group and ACEI group were no obvious thickening of myocardial fibers,no significant inter-band gap,subendocar only have a small amount of collagen fiber and reduce the content of Hydro in myocardial tissue(P<0.05),but the Jie Du Tong Luo Fang each dose group compared with ACEI group have no significant difference(P>0.05).These results suggest that Jie Du Tong Luo Fang each dose group and ACEI group have similar effects on reversing or controlling myocardial fibrosis.(2)Cmopared with normal control group,the Jie Du Tong Luo Fang each dose group and ACEI group had a significant effect on reducing the content of tTG in myocardial tissue(P<0.05),but the Jie Du Tong Luo Fang each dose group compared with ACEI groupb have no significant difference on the content of tTG protein in myocardial tissue(P>0.05).The results indicated that the effect of tTG on the control of myocardial fibrosis was related to the decrease of the expression of.Conclusion:Jie Du Tong Luo Fang may improve the MF in rats by lowering the expression of tTG in the MF rat. |
