Objective:The Lewis lung cancer mouse was used as an experimental animal model to detect the role of vascular endothelial growth factor(VEGF),cyclooxygenase-2(COX-2)and CD34 in tumor tissues of Lewis lung cancer by immunohistochemistry.Mechanism,explore the anti-cancer mechanism of lung cancer I,provide a reliable scientific basis for its clinical application.Method:An animal model of 48 SPF C57 Lewis lung cancer mice was established and randomly divided into four groups: tumor-bearing control group,Chinese medicine group(lung cancer group I),DDP group(cisplatin),and Chinese medicine combined with DDP group.The tumor-bearing control group received intragastric administration of NaCl(1 time per day)and intraperitoneal injection of NaCl(3 times per day);the Chinese medicine group was intragastrically administrated with lung cancer I prescription(1 time per day)and intraperitoneal injection of NaCl(3 times per day).The DDP group was intragastrically instilled with NaCl(1 time per day)and intraperitoneal injection of DDP(3 times per day);Chinese medicine combined with DDP group was intragastrically administered with lung cancer I prescription(1 time per day),and DDP was intraperitoneally injected DDP(3 times per day).On the thirteenth day,the model mice were sacrificed by spinal dislocation and the tumor tissues were removed and weighed.The tumor tissues were fixed,followed by wax,sectioning,staining,and histopathological examination was performed.Immunohistochemical SP method was used to examine the expression of vascular endothelial growth factor,cyclooxygenase-2,and microvessel density in tumor tissues.And use SPSS 22.0 software for statistical analysis of data.Result:1.Measurement results of tumor weight and tumor inhibition rate1.1 Tumor weight:The results showed that compared with the control group,the average tumor weight of the Chinese medicine group was statistically significant(P<0.05),and the average tumor weight of the DDP group was lower than that of the control group(P<0.01).The mean tumor weight of Chinese herbs combined with DDP group was significantly lower than that of the control group(P<0.01);with the Chinese Herbal Medicine group as the comparison object,the average tumor weight of the DDP group was lower than that of the Chinese medicine group.The difference was statistically significant(P<0.05),Chinese herbs combined with DDP group's average tumor weight was significantly lower than the Chinese medicine group(P<0.01);with DDP group as the comparison object,the average tumor weight of Chinese herbs combined with DDP group was lower than that of DDP group(P<0.01).1.2 Determination of tumor inhibition rate:The inhibitory rate of Chinese medicine combined with DDP group was 41.8%,and the curative effect was the best in the tumor suppressing rate.The tumor inhibiting rate in DDP group was 26.6%,and the curative effect was slightly weak in the tumor suppressing rate;the inhibitory rate of Chinese medicine group was 14.7%.The cancer rate is not as effective.2.Results of positive expression of vascular endothelial growth factor(VEGF):The positive expression of vascular endothelial growth factor is based on its average optical density value.The results showed that the average optical density of vascular endothelial growth factor was: tumor control group(0.1453 ± 0.0036);Chinese medicine group(0.1383±0.0035);DDP group(0.1252±0.0052);Chinesemedicine combined DDP group(0.1123 ± 0.0065)The average optical density of VEGF in the TCM group was lower than that in the tumor-bearing control group(P<0.05);the mean optical density of VEGF in the DDP group was lower than that in the tumor-bearing control group(P<0.01).The mean optical density of VEGF in Chinese medicine combined with DDP group was lower than that in tumor-bearing control group(P<0.01);the average optical density of VEGF in DDP group was lower than that in Chinese medicine group(P<0.01).The average optical density of VEGF in Chinese medicine combined with DDP group was lower than that in Chinese medicine group(P<0.01);the average optical density of VEGF in Chinese medicine combined with DDP group was lower than that in DDP group(P<0.01).3.Results of positive expression of cyclooxygenase-2(COX-2)The positive expression of cyclooxygenase-2 was based on its average optical density value.The results showed that the average optical density values of cyclooxygenase-2 were: tumor-bearing control group(0.1456 ± 0.0060);Chinese medicine group(0.1361±0.0041);DDP group(0.1119±0.0106);Chinese medicine combined with DDP group(0.1028±0.0082),the mean optical density of COX-2 in Chinese Herbal Medicine group was lower than that in tumor-bearing control group(P<0.01);the average optical density of COX-2 in DDP group was lower than that in tumor-bearing control group.Significance(P<0.01);The average optical density of COX-2 in Chinese herbs combined with DDP group was lower than that in tumor-bearing control group(P<0.01);The average optical density of COX-2 in DDP group was lower than that in Chinese medicine There was a statistically significant difference between the groups(P<0.01);the average optical density of COX-2 in Chinese herbs combined with DDP group was lower than that in Chinese medicine group(P<0.01);the positive rate of COX-2 in Chinese herbs combined with DDP group was statistically significant.The optical density value was lower than the DDP group but the difference was not statistically significant(P>0.05).4.Positive expression of CD34The positive expression of CD34 was based on the average optical density value.The results showed that the average optical density of microvessel density was:tumor-bearing control group(0.1481±0.0063);Chinese medicine group(0.1353±0.0023);DDP group(0.1132±0.0077);Chinese medicine combined with DDP group(0.1041±0.0067);The average optical density of CD34 in the group was significantly lower than that in the tumor-bearing control group(P<0.01);the average optical density of CD34 in the DDP group was lower than that in the tumor-bearing control group(P<0.01);The average optical density of CD34 in the combined DDP group was lower than that in the tumor control group(P<0.01).The average optical density of CD34 in the DDP group was lower than that in the Chinese medicine group(P<0.01).The average optical density of CD34 in Chinese medicine combined with DDP group was lower than that in Chinese medicine group(P<0.01);the average optical density of CD34 in Chinese medicine combined with DDP group was lower than that in DDP group(P<0.05).5.Observe the lung cancer tissue section:Compared with the tumor-bearing control group,the remaining three groups had increased areas of necrosis,a wide area of necrosis,focal or platy necrosis,and significant cell edema,and the density became lower.The cell atypicality is relatively reduced.Comparing the other three groups,the necrosis area of the DDP group was larger in the Chinese herbal medicine group,and the area of the necrosis area in the DDP group was wider than that of the Chinese medicine group.In Conclusion:1.Lung cancer I can inhibit the growth of tumors by reducing the expression of VEGF,COX-2,and CD34 in Lewis lung cancer mice and inhibiting the formation of blood vessels in tumor tissues.2.In combination with the chemotherapeutic drug DDP,Lung Cancer No.I is most effective in inhibiting angiogenesis in tumor tissues and can effectively inhibit tumor growth.
