Objective : SQSW tablet is an empirical prescription for the old Chinese Medicine,Clinical and preliminary experimental studies have confirmed its role in treating hypertension and delaying hypertension renal damage.In this study,we observed the effects of SQSW tablets on c AMP,cyclic c GMP and c AMP/c GMP in plasma of SHR rats.Based on the related research and the theory of yin and Yang nucleotide,explore the tablets SQSW in the treatment of hypertension possible.Overexpression of Ang II in kidney is one of the causes of renal damage.AC-c AMP pathway is Ang II signaling system in kidney.To observe the effect of SQSW tablet on AC and c AMP in the kidney of SHR,and to analyze the possible causes of SQSW tablet to delay the damage of hypertensive kidney.Method:In this study,SHR rats were used as a model of low,medium and high dose groups,and a blank control group,a Compound Salvia Tablet group,a Kato Pury group and a normal WKY rat group.Five weeks of continuous administration,collecte orbital blood samples in ever week.The content of c AMP and c GMP in plasma was detected by ELISA method,and the ratio of c AMP/c GMP was calculated.Renal tissues were extracted after the last dose and 7d.The contents of AC,c AMP and Ang II in renal tissue were detected by ELISA.The statistical results were expressed with mean standard deviation((?)ąs).Single factor analysis of variance was used,every two group were compared with SNK-q test.The difference was statistically significant with the difference of P<0.05.Result:(1)During the period of administration,the plasma c AMP level in the blank control group and the drug delivery group was higher than that in the normal control group(P< 0.05);There was no significant difference between the administration and the blank control group in 1d;In 7d,14 d,21d,28 d and 35 d the content of c AMP in the high-dose group was lower than the blank control group,and the difference was statistically significant(P<0.05);In 21 d,28d and 35 d the content of c AMP in plasma of in medium and low dose group was lower than blank control group,the difference was statistically significant(P < 0.05).(2)During the period of administration,the plasma c GMP level in the blank control group and the drug delivery group was lower than that in the normal control group(P< 0.05);There was no significant difference between the administration and the blank control group.in 1d;In 28 d,the content of c GMP in high dose group was higher than that in blank control group,the difference was statistically significant(P<0.05);In35d,the content of c GMP in every dose group,was higher than that in blank control group,the difference was statistically significant(P<0.05).(3)During the period of administration,the c AMP/c GMP in the blank control group and the drug delivery group was higher than that in the normal control group(P<0.05);There was no significant difference between the administration and the blank control group in 1d;In 14 d,the c AMP/c GMP in the low and medium dose group was lower than the blank control group,and the difference was statistically significant(P<0.05);The c AMP/c GMP in the low,medium and high dose groups of 21 d,28d and35 d were lower blank control group,the difference was statistically significant(P<0.05).(4)At the end of the last drug delivery and 7d,the AC in the renal tissue was higher in the blank control group and the drug groups than in the normal control group,with statistical significance(P<0.05);The end of the last administration was compared with the blank control group,and the AC decreased in the renal tissue and was statistically significant(P<0.05);the drug group was stopped with 7d,and the drug groups were compared with the blank control.Compared with the control group,AC in renal tissue was still decreased,with statistical significance(P<0.05).(5)At the end of the last administration and 7d,the c AMP in the renal tissue was significantly higher in the blank control group and the drug group than in the normalcontrol group(P<0.05),and the cAMP in the renal tissue at the end of the last administration group was significantly lower than that in the blank control group(P <0.05);the withdrawal of 7D and the c AM in the renal tissue of each administration group were c AM.P was lower than that in blank control group,but not statistically significant(P > 0.05).(6)At the end of the last administration and 7d,the Ang II in the renal tissue of the blank control group and the drug groups was higher than that in the normal control group.The Ang II in the renal tissue of each dose group of Sanqi Shouwu tablet was lower than that of the blank control group at the end of the last Administration(P<0.05),and the dosage of Ang II in each dose group of Sanqi Shouwu tablet was still higher than that of the blank control group.There were statistical significance in the group(P<0.05).Conclusion:1.he plasma levels of c AMP in SHR rats increased,c GMP decreased,the ratio of c AMP/c GMP increased,and there was a disorder of cyclic nucleotides.SQSW tablet could reduce the c AMP in the plasma of SHR rats,increase the c GMP,reduce the c AMP/c GMP ratio,correct the disorder of cyclic nucleotide and balance the c AMP/c GMP.2.The increase of AC,c AMP and Ang II in the kidneys of SHR rats can reduce the AC,c AMP and Ang II of the kidney of SHR rats,which may be a mechanism of renal protection.
