Ovarian cancer is one of the three most common malignant tumors in the female reproductive organs.Although morbidity ranks third in gynecologic malignant tumor,the mortality was the highest.Epithelial ovarian cancer is the most common ovarian malignancy in ovarian epithelial neoplasms.It was found to be late because of the lack of early molecular biological diagnostic indicators and distinctive clinical manifestations,early lesions are not easy to detect.Despite ovarian cancer may receive clinically relieved by early active acceptance of interval debulking surgery reduction and platinum drugs combined paclitaxel(TP)combination chemotherapy scheme,it is known as a first-line treatment for ovarian cancer.Chemotherapy drug resistance and postoperative easy to recurrent ovarian cancer mortality is still first in gynecologic malignant tumors.It becomes a serious cancer that threatens women's lives and health.So the research on the pathogenesis and early molecular biological diagnosis index of ovarian cancer has been the focus of research on gynecological tumors at domestic and overseas.Epithelial mesenchymal Transformation(EMT)is epithelial cells transformed into mesenchymal cells to obtain the peculiarity of mesenchymal cells.It is participated in embryonic development,tissue repair,wound healing and neoplasm metastasis.E-cadherin is the significant epithelial cells transcriptional inhibitor factor in EMT process.It is a transmembrane glycoprotein of ca-dependent.The main function of E-cadherin is to mediate the adhesion reaction between homogeneous cells.It's prevailingly embodied in the cytomembrane.Zinc finger structural protein super family Slug,also named Snail2.It consists of a highly conserved C-terminal hydroxyl group containing Zinc finger domain and an N-terminal amino terminal.Slug is combined with E-box of E-cadherin start up element.and Slug is the suppressor factor of E-cadherin promoter can reduce adhesion force between cells by specificity inhibition of epithelial cell fenotyp constituent E-cadherin.It is the direct inhibitory factor regulating the transcription of E-cadherin.ObjectiveThere have been no reports in regard to combining expression of Slug and E-cadherin in EOC.In this study immunohistochemical SP and qRT-PCR were used to detect the protein positive expression rates and mRNA expressions of Slug and E-cadherin in different ovarian tissues.Analyze the relationship among the protein positive expression rates of the two factors and different clinical pathological features in ovarian epithelial carcinoma tissues,and the correlation of mRNA relative transcript level of the two factors in epithelial ovarian carcinoma tissues was also analyed.In order to discuss the mechanism and interaction of Slug and E-cadherin in ovarian cancer development and metastasis,to explore whether the two could be a potential diagnostic marker for EOC.Materials and Methods1.Clinical DataA total of 104 patients were collected From February 2015 to March 2017 who were hospitalized in parallel to remove the ovarian tissues in the third clinical college of Zhengzhou university.There were 20 normal ovarian tissues,The normal ovary tissues derived from the adnexa have been surgical resected because of endometrial carcinoma,cervical cancer and other sickness,30 cases of benign ovarian tumor epithelial tissues and 54 cases of epithelial ovarian carcinoma tissues.Postoperative pathology confirmed that there was no lesion in the ovary.All patients were first found,and all patients have not been any radiotherapy and chemotherapy before surgery.All specimens were pathologically diagnosed by the pathologists of our hospital,and get the agreement of subjects and the Ethics Committee.2.Experimental Methods(1)Immunohistochemical SP was used to test the protein positive expression rates of Slug and E-cadherin in the normal ovarian tissues,ovarian benign epithelial tumor tissues and epithelial ovarian carcinoma tissues.The relationship between the protein positive expression rates of the two divisors and the clinicopathological features in epithelial ovarian cancer was analyzed.(2)qRT-PCR was used to examine Slug and E-cadherin mRNA expression level in the normal ovarian tissues,ovarian benign epithelial tumor tissues and epithelial ovarian carcinoma tissues.and the correlation between the mRNA expressions level of two factors in epithelial ovarian cancer were analyed.3.Statistical methodsStatistic analysis by using SPSS21.0.The counting data is expressed as a percentage(%).The comparison between the counting data was conducted with the?~2test.Quantitative data is expressed in x±s.LSD-t was used to compare the quantitative data between the two groups.The quantitative data of the three groups were analyzed by One-Way ANOVA.Pearson correlation analysis was used in correlation analysis.Significant level?=0.05,P<0.05 has statistical significance.When compared between two groups,test level??=?/comparison times=0.0167.Result1.The protein positive expression rates of Slug and E-cadherin in different ovary tissues.The protein positive expression rates of Slug in normal ovarian tissues,ovarian benign epithelial tumor tissues and epithelial ovarian carcinoma tissues were5.0%,43.3%and 88.9%.The comparison between the two groups is shown:The malignant ovarian group was higher than the benign ovarian tumor group and normal ovarian tissue group,and the difference was statistically significant(P<0.05).The benign ovarian tumor group was higher than the normal ovarian tissue group,The difference was statistically significant(P<0.05).The positive expression rate of E-cadherin in normal ovarian tissues,ovarian benign epithelial tumor tissues and epithelial ovarian cancer tissues were 85.0%,46.7%and 22.2%.Between the two groups,the malignant ovarian group was lower than the benign ovarian tumor group and the normal ovarian tissue group,and the differences were statistically significant(P<0.05).The benign ovarian tumor group was lower than the normal ovarian tissue group,and the difference was statistically significant(P<0.05).2.The relationship between the positive expression rate of Slug and E-cadherin protein and the clinicopathological features in epithelial ovarian cancer.The protein positive expression rates of Slug in the epithelial ovarian cancer tissues with FIGO I-II and III-IV were 75%and 97.1%,The protein positive expression rate of Slug in epithelial ovarian cancer with advanced-stage was higher than that patients with early-stage with the pathological staging,The protein positive expression rate of Slug increased with the pathological stage of operation increased,The difference was statistically significant(P<0.05).The positive expression rate of Slug protein with lymph node metastasis and with no lymph node metastasiswere 100.0%and 71.4%,The positive expression rate of Slug protein with lymph node metastasis was higher than with no lymph node metastasis,The difference was statistically significant(P<0.05).The positive expression rate of Slug protein with ascites and no ascites were 97.1%and 73.7%,The positive expression rate of Slug protein with ascites was higher than that without ascites in ovarian cancer,The difference was statistically significant(P<0.05).It was not related to patientage,histological type and histological classification(P<0.05).The protein positive expression rates of E-cadherin in the epithelial ovarian cancer tissues with FIGO I-II and III-IV were 40%and 11.8%,The protein positive expression rate of E-cadherin in epithelial ovarian cancer with advanced-stage was inferior to that patients with early-stage with the pathological staging,The protein positive expression rate of E-cadherin was decreased with the pathological stage of operation increased,The difference was statistically significant(P<0.05).The positive expression rate of E-cadherin protein with lymph node metastasis and with no lymph node metastasiswere 9.1%and 42.9%,The positive expression rate of E-cadherin protein with lymph node metastasis was inferior to with no lymph node metastasis,The difference was statistically significant(P<0.05).The positive expression rate of E-cadherin protein with ascites and no ascites were11.4%and 42.1%,The positive expression rate of E-cadherin protein with ascites was inferior to that without ascites in ovarian cancer,The difference was statistically significant(P<0.05).It was not related to patientage,histological type and histological classification(P<0.05).3.The mRNA expression levels of Slug and E-cadherin in different ovarian tissues.The mRNA relative expression levels of Slug in the normal ovarian group,benign epithelial ovarian tumor group and epithelial ovarian cancer group were0.480±0.279,1.262±0.551 and 2.346±0.676.The mRNA expression level of in epithelial ovarian cancer group was higher than those of in benign epithelial tumor tissue group and normal ovary group,The difference were statistically significant(P<0.05).The mRNA expression level of Slug in benign epithelial ovarian tumor group was higher than that of normal ovary group,The difference between the two groups was statistically significant(P<0.05).The mRNA relative expression of Slug Between three groups was statistically significant use One-Way ANOVA(P<0.05).The mRNA relative expression of E-cadherin in the normal ovary group,benign epithelial ovarian tumor group and epithelial ovarian cancer group were 2.342±0.503,1.139±0.449 and 0.490±0.315.The m RNA expression level of E-cadherin in epithelial ovarian cancer group was lower than that in benign epithelial ovarian tumor group and normal ovarian group,The difference was statistically significant(P<0.05).The mRNA expression level of E-cadherin in benign epithelial tumor group was lower than that in normal ovarian group,The difference between the two groups was statistically significant(P<0.05).The m RNA expression relative of E-cadherin among the three groups was statistically significant employ One-Way ANOVA(P<0.05).4.The correlation analysis of the mRNA expression levels of Slug and E-cadherin in epithelial ovarian carcinoma tissues.The mRNA expression levels of Slug and E-cadherin in ovarian epithelial carcinoma tissues were negative correlationget through Pearson correlation analysis,and the difference was statistically significant(r=-0.276,P<0.05).Conclusion1.High expression of Slug and low expression of E-cadherin protein in epithetlial ovarian cancertissues,suggesting that both factors may play an important role in the development and metastasis of ovarian cancer,They can be used as a reference for determining the malignancy and biological behavior of ovarian cancer.2.The mRNA expression levels of Slug and E-cadherin in epithelial ovarian cancer tissues were negatively correlated.It is suggested that there may be mechanism of mutual regulation between Slug and E-cadherin in the development and metastasis of ovarian cancer.Slug as a direct transcription inhibitor of E-cadherin,induce the occurrence of EMT by inhibiting the expression of E-cadherin and lead to the occurrence and metastasis of the tumor.
