Effect Of Yiqi Jiedu Method On T The Growth And Expression Of Related Proteins In Subcutaneous Xenografts Of Nude Mice

Objective:To investigate the effect of Yiqi Jiedu Decoction on the proliferation of tumor cells and the mechanism of apoptosis induced by anti-tumor detoxification by using different concentrations of gradient gaseous detoxification solution in nude mice bearing the gastric cancer cell line of human gastric cancer MGC-803 cell line Experimental study,for clinical application to provide a certain experimental basis.Methods:1.To establish the lung cancer cell line MGC-803 in nude mice,4 to 6 weeks old and15 to 18 g BALB/c male nude mice were selected.The MGC-803 gastric cancer cells were treated with PBS Liquid to suspend,adjust the cell concentration to 1�10~7/ml,0.2 ml/only inoculated in 4 nude mice on the left armpit.When the tumor was increased to the maximum diameter of about 1cm,the tumor pieces were stripped and the tumor pieces were cut into 100mm dishes and cut into 2 mm�2 mm�2 mm tumor tissue pieces.The left armpits were implanted in the left side of the nude mice,Construction of tumor-bearing nude mice.2.Experimental group and drug intervention The nude mice were divided into 6 groups according to the random number table,the first group is saline group,the second group is high-dose detoxification group,the third group is qi decoction in the middle dose group,Four groups is low-dose detoxification side,the fifth group is 5-Fu group,the sixth group is traditional Chinese medicine dose+5-Fu group,each group of 10.The infusion dose was 0.1 ml/10 g nude mice.3.The results analyzed the weight of the nude mice was recorded during the experiment.The tumor length was calculated and the tumor growth curve was drawn.The tumor growth curve was calculated.The effect of Yiqi Jiedu Decoction on TNF-?in nude mice was detected by ELISA.The expression of Survivin in each tumor was detected by Western Blot,and the expression of Survivin was detected by Western Blot.Livin,Bcl-2,Bax,Caspase-3 and other protein expression.Results:1.The weight gain of nude mice and the weight gain curve of nude mice:There was no significant difference between the normal saline group and the traditional Chinese medicine group(P>0.05)(P<0.01).Compared with the traditional Chinese medicine group,the 5-Fu group had significant difference(P<0.05).2.The tumor volume and growth curve:compared with the normal saline group,low dose of Chinese medicine group was not statistically significant(P>0.05),Chinese medicine dose group was statistically significant(P<0.05),there was significant dose 5-Fu group high dose of Chinese medicine group,5-Fu group,Chinese herbal medicine(P<0.01).Compared with the 5-Fu group,Chinese herbal medicine and low dose group were statistically significant(P<0.01),high dose of Chinese medicine group was statistically significant(P<0.05),no statistical significance with Chinese medicine dose combined with 5-Fu group(P>0.05).3.The tumor weight and tumor inhibition rate:compared with the normal saline group,Chinese medicine high dose group,there was significant dose combination group 5-Fu 5-Fu group,Chinese medicine(P<0.01),low dose of Chinese medicine group was not statistically significant(P>0.05).Compared with the 5-Fu group,the middle and low dose groups of Chinese medicine had significant statistical significance(P<0.01),the high dose group of Chinese medicine was statistically significant(P<0.05),and there was no statistical significance(P>0.05)in the combination of Chinese medicine with the dose of 5-Fu.4.The levels of TNF-?in ELISA were significantly higher than those in the control group(P<0.01).Compared with the normal saline group,the high dose,medium and low dose group,5-Fu group and traditional Chinese medicine were significantly higher than those in the 5-Fu group(P<0.01).Compared with the 5-Fu group,the low-dose group of Chinese medicine had significant statistical significance(P<0.01),and there was no statistical significance(P>0.05)between the high and middle group and the traditional Chinese medicine combined dose 5-Fu group.5.Western Blot experimental results(1)Survivin protein expression:compared with the normal saline group,the high,medium and low dose groups of Chinese medicine,5-Fu group,Chinese medicine combined dose of 5-Fu group has significant statistical significance(P<0.01).Compared with the 5-Fu group,the middle and low dose groups of Chinese medicine had significant statistical significance(P<0.01),the high dose group of Chinese medicine was statistically significant(P<0.05),and there was no statistical significance(P>0.05)in the combination of Chinese medicine with the dose of 5-Fu.(2)Livin protein expression:compared with the normal saline group,the high,medium and low dose groups of Chinese medicine,5-Fu group,Chinese medicine combined dose of 5-Fu group has significant statistical significance(P<0.01).Compared with the 5-Fu group,the high,medium and low dose groups of Chinese medicine had significant statistical significance(P<0.01),and there was no statistical significance(P>0.05)in the combination of Chinese medicine with the dose of5-Fu.(3)Bcl-2 protein expression:compared with the normal saline group,the high,medium and low dose groups of Chinese medicine,5-Fu group,Chinese medicine combined dose of 5-Fu group has significant statistical significance(P<0.01).Compared with the 5-Fu group,there was significant low dose of Chinese medicine group(P<0.01),Chinese medicine high dose group was statistically significant(P<0.05),no statistical significance with Chinese medicine dose combined with 5-Fu group(P>0.05).(4)The expression of Bax protein:compared with the normal saline group,there was significant dose combination group 5-Fu 5-Fu group(P<0.01),Chinese medicine,Chinese medicine high dose group was statistically significant(P<0.05),low dose of Chinese medicine group was not statistically significant(P>0.05).Compared with the 5-Fu group,Chinese herbal medicine and low dose group were statistically significant(P<0.01),high dose of Chinese medicine group was statistically significant(P<0.05),no statistical significance with Chinese medicine dose combined with 5-Fu group(P>0.05).(5)Bax/Bcl-2:compared with the normal saline group,the high and middle dose groups of Chinese medicine,the 5-Fu group and the medium dose of Chinese medicine combined with the 5-Fu group had significant statistical significance(P<0.01),and there was no statistical significance in the low dose group(P>0.01).Compared with the 5-Fu group,the high,medium and low dose groups of Chinese medicine had significant statistical significance(P<0.01),and there was no statistical significance(P>0.05)in the combination of Chinese medicine with the dose of 5-Fu.(6)The expression of Caspase-3 protein:compared with saline group,Chinese medicine high dose group,there was significant dose combination group 5-Fu 5-Fu group,Chinese medicine(P<0.01),low dose of Chinese medicine group was not statistically significant(P>0.01).Compared with the5-Fu group,the high,medium and low dose groups of Chinese medicine had significant statistical significance(P<0.01),and there was no statistical significance(P>0.05)in the combination of Chinese medicine with the dose of 5-Fu.Conclusion:1.Through the overall observation of nude mice and tumor measurements,indicating that Yiqi Jiedu method does not affect the weight of nude mice under the premise of inhibition of tumor growth.2.Yiqi Jiedu method can reduce the content of TNF-?.3.Yiqi Jiedu method can reduce the expression of apoptotic protein,while upregulation to promote the expression of apoptotic protein.4.The effect of high and middle dose group was better than that of low dose group.
