objectives The aim of the study was to explore the effect of dexmedetomidine on the proliferation and apoptosis of neuroblas toma.Methods 1?The SH-SY5 Y cells were respectively treated wit h 0?0.1uM?1uM?10u M dexmedetomidine and screened optimal co ncentration;dexmedetomidine dealed with the SH-SY5 Y cells about0?6h?12h?24h respectively and screened optimal time;The SH-S Y5 Y cells were respectively treated with 0?100uM?150uM?200uM yohimbine and screened optimal concentration;yohimbine dealed w ith the SH-SY5 Y cells about 0?6h?12h?24h respectively and scre ened optimal time.Cell proliferation was evaluated by MTT assays.2?In optimal time,the SH-SY5 Y cells,dividing into control group?dexmedetomidine group and yohimbine group were incubated with o ptimal concentration.The effect of dexmedetomidine and yohimbine on apoptosis was assessed by flow cytometry.3?q-PCR analyzed t he mRNA expression of ?2-adrenergic receptor of SH-SY5 Y cells.Results MTT: Cell proliferation rate increased as dexmedetomidine's concentration increased and dexmedetomidine's optimal con centration was 1uM;with time increased,Cell proliferation rate incre ased and dexmedetomidine's optimal time was 12h;Cell proliferation rate decreased as yohimbine's concentration increased and yohimbi ne's optimal concentration was 200uM;with time increased,Cell pro liferation rate decreased and yohimbine's optimal time was 12h;FC M:The apoptosis rate of dexmedetomidine group was lower than th at of control group;The apoptosis rate of yohimbine group was hi gher than that of control group.q-PCR:SH-SY5 Y cells expressed ?2-adrenergic receptors.Conclution Dexmedetomidine could enhance SH-SY5 Y proliferation and inhibited its apoptosis.SH-SY5 Y cells expressed ?2-adrenergic receptors.
