The Research About The Implantation Metastasis Rate Of Overflowing Human Lung Cancer Cells Plant In The Nude Mice After Needle Biopsy Of Lung

Objective: Through a research to prove that there are lung cancer cells in the washing fluids of the needle hole of the visceral pleura,the needle tip and the tissue strips in the stylet and the tumor will be formed after these washing fluids planted under the skins of the nude mice.Meanwhile,the tumor forming rate will be obtained and the nude mice being used whether sensitivity for this experiment will be verified.Therefore,clinic doctors will make an appropriate choice when they decide to give patients needle biopsy of lung or not.Methods:(1)40 fresh intact human lung lobes were excised with the thoracotomy pulmonary lobectomy.These lung lobes from the thoracic surgery department of the Hu-Nan Cancer Hospital were acquired without radiotherapy and chemotherapy and pathological diagnosis showed lung cancer;(2)40 BALB/CA-NU,4-8 weeks of age,weight 18-22 g,half male and half female nude mice were fed in grade SPF aseptic room of animal experiment center of Hu-Nan Cancer Hospital;(3)The above 30 lung lobes were punctured by TSK semi-automatic biopsy needles 5 times,the needle hole of the visceral pleura?the needle tip and the tissue strips in the stylet were washed by a little normal saline respectively and repeatedly.3 pipes washing fluid were collected and been concentrated to0.2ml;(4)The tumor bodies of remaining 10 lung lobes were incised and10 pieces of mung size of tumor bodies were taken;(5)40 nude mice were separated to 2 parts,one part included 30 mice,3 pipes washing fluid were planted at 3 different points under their skins.After planting,3 pipes were washed respectively and 3 washing fluid of pipes were collected and then the slide review been done to find cancer cells;Another part included 10 mice and the mung size tumor bodies were planted under their skins;(6)All mice were fed 30 days and then been executed to exam whether tumor bodies been formed and the tumor bodies been formed were taken to do paraffin section for affirming had cancer or not.The tumor bodies been formed and original patients were checked to affirm whether the type of cancer and the differentiated degree were same between them.The end,the tumor formation rate was obtained.Result:(1)Tumor bodies were not formed at the washing fluid planting points of the needle tip and the tissue strips in the stylet;(2)1 tumor body was formed at the point of the needle hole of the visceral pleura,the formation rate was 3.3%,the cancer type was adenocarcinoma.;(3)Cancer cells in the washing fluid were found;(4)7 tumor bodies were formed under skin of mice planted tumor bodies,the formation rate was70%,the cancers type all were adenocarcinoma Conclusion:(1)There are lung cancer cells in the washing fluids of the needle hole of the visceral pleura,the needle tip and the tissue strips in the stylet.Therefore,there is a risk of implantation metastasis after doing needle biopsy of lung;(2)There is a low tumor forming rate after the washing fluid of the needle hole of the visceral pleura planted under the skins of the nude mice(3.3%);(3)Can not completely exclude the tumor forming ability of the washing fluids of the needle tip and the tissue strips in the stylet after there washing fluids planted under the skins of the nude mice;(3)All formation tumors of this experiment are adenocarcinoma,indicating the adenocarcinoma is more likely to occur implantation metastasis.We can enlarge the number of specimens and researching deeply in the future.(5)The BALB/CA-NU nude mouse is sensitive and appropriate for this experiment.
