Objective:To systematically evaluate the association between XRCC1 Arg399Gln polymorphism and the risk of digestive system cancer in Chinese population,so as to provide a more comprehensive and reliable theoretical and clinical basis.Methods:All studies related to XRCC1 Arg399Gln polymorphism and the risk of digestive system cancer in Chinese population were searched in PubMed,Medline,Web of Science,Embase,CNKI,VIP,CBM and WanFang databases.The pooled ORs with 95%CIs were calculated by Revman 5.2 and Stata 12.0 software.Publication bias and sensitivity were also evaluated at the same time.Results:56 studies met our inclusion criteria about the association between XRCCl Arg399Gln polymorphism and risk of digestive system cancer in Chinese population.Among the included studies,14 studies were about esophageal cancer,12 studies were about gastric cancer,13 studies were about colorectal cancer,13 studies were about hepatocellular carcinoma,4 studies were about pancreatic cancer.54 published case-control studies with 14137 digestive system cancer patients and 19858 controls were included in this meta-analysis.The analysis showed significant interactions between XRCC1 Arg399Gln polymorphism and hepatocellular carcinoma under five genetic models[(Gln vs Arg:OR= 1.33,95%CI:1.16-1.54,P<0.001),(Gln/Gln vs Arg/Arg:OR= 1.47,95%CI:1.24-1.74,P<0.001),(Arg/Gln vs Arg/Arg:OR = 1.48,95%CI:1.19-1.84,P<0.001),(Gln/Gln vs Arg/Arg+Arg/Gln:OR = 1.26,95%CI:1.08-1.48,P = 0.003),(Arg/Gln+Gln/Gln vs Arg/Arg:OR = 1.49,95%CI:1.21-1.83,P<0.001)].However,there was no correlation between XRCC1 Arg399Gln polymorphism and the risk of esophageal cancer,gastric cancer,colorectal cancer and pancreatic cancer.Conclusion:The present meta-analysis indicates that XRCC1 Arg399Gln polymorphism may be associated with the risk of hepatocellular carcinoma.However,there was no correlation between XRCC1 Arg399Gln polymorphism and the risk of esophageal cancer,gastric cancer,colorectal cancer and pancreatic cancer.
