Effect Of SiRNA DAD1 And G-Rh2 On Proliferation And Apoptosis Of Human Bladder Cancer BIU-87 Cell | Posted on:2018-08-24 | Degree:Master | Type:Thesis | Country:China | Candidate:X Liu | Full Text:PDF | GTID:2334330542466699 | Subject:Clinical Medicine | Abstract/Summary: | PDF Full Text Request | Objective: To observe the effect of DAD1 gene silencing on the in vitro anti-bladder cancer efficiency of ginsenoside Rh2 and to explore the feasibility and synergistic anti-tumor effect of herbal extract ginsenoside Rh2 combined with RNAi technology,providing laboratory support for the comprehensive therapy of tumors.Method: Bladder carcinoma cell line BIU-87 was used as the subjects.BIU-87 cells were treated with different concentrations of ginsenoside Rh2,and proliferation ability of the cells was detected by MTT assay to screen the optimum conditions for ginsenoside Rh2;ginsenoside Rh2 was then combined with siRNA-DAD1 transfection to intervene the BIU-87 cells;proliferation of BIU-87 cells after combined intervention was measured by MTT assay;expression level of DAD1 mRNA was determined by real-time fluorescent quantitative PCR;expression level of DAD1 protein was detected by Western blot;apoptosis level of BIU-87 was measured by flow cytometry;and invasion ability of these cells was detected by Transwell assay.Results:Proliferation ability of bladder carcinoma cell line BIU-87 was inhibited to certain degrees by different concentrations of ginsenoside Rh2 at different intervention time(P<0.05);when treated with ginsenoside Rh2 at 40umol/L combined with siRNA-DAD1,MTT assay showed that the proliferation of BIU-87 cells was decreased(P<0.05);fluorescence quantitative PCR detection indicated that the expression of DAD1 mRNA was reduced in BIU-87 cells(P<0.05);Western blotting showed that the expression level of DAD1 protein was decreased(P<0.05);flow cytometry indicated that apoptosis of BIU-87 cells was increased(P<0.05);and Transwell assay demonstrated that the invasion ability of BIU-87 cells was significantly decreased(P<0.05).Conclusion:1.Ginsenoside Rh2 of certain concentrations inhibits the proliferation of bladder carcinoma cell line BIU-87.2.Ginsenoside Rh2 combined with siRNA-DAD1 significantly inhibits the expression of DAD1,induce the apoptosis and inhibit the invasion of bladder carcinoma cell line BIU-87. | Keywords/Search Tags: | DAD1, Bladder cancer, Ginsenoside Rh2, Cell proliferation, Cell invasion | PDF Full Text Request | Related items |
