Screen And Study On Anti Fungal In Vitro Effective Parts Of Gelsemium

Purpose The purpose of this study is to explore the antifungal effect of Gelsemium extract in vitro,and explore specific parts of Gelsemium extract in vitro antifungal.Methods The extraction of Gelsemium water extract and ethanol extract of Gelsemium powder with ultra pure water and 95%ethanol;the ethanol extract respectively with petroleum ether,ethyl acetate and n-butanol,get the corresponding parts of the powder;by using the HPLC method of gelsemine,Gelsemium elegans alkali C,Koumine,hexa gelsemicine,gelsevirine,evergreen gelsemine and alkali Gelsemium elegans B seven standard,and the detection of alcohol extract of Gelsemium alkaloids extracts;43 strains of superficial fungal detection and the extraction of components of Gelsemium extract on clinical collected and Chinese academy skin disease Research Institute of purchasing by micro broth dilution method of Trichophyton rubrum,Trichophyton,Microsporum,Epidermophyton and Candida albicans antifungal activity of 5 strains in vitro;detection of Gelsemium ethanol extract and its fractions of Aspergillus fungus in vitro by agar dilution method.Result 1.Ge water extract freeze-dried powder weight is 10.532g,the yield was 10.5%;the ethanol extract of Gelsemium powder weight is 4.752g,the yield was 4.8%.Alcohol extract of Gelsemium after extraction of petroleum ether fraction 0.566g,a rate of 11.8%;the 1.885g of ethyl acetate,the yield was 39.7%;n-butanol fraction 1.583g,a rate of 33.3%;part 0.563g,the yield was 11.8%,ethyl acetate was the highest.2.Seven kinds of index of n-butanol fraction of alkaloids of Gelsemium alcohol extract:gelsemine and Hu Manteng base C,Koumine,hexa gelsemicine,gelsevirine,sempervirine and huperzine B the highest content of Gelsemium elegans.Gelsemine alcohol position of petroleum ether fraction and ethyl acetate from no alkaloid content is higher.3.The identification results of 43 strains of fungi tested as follows:Trichophyton rubrum 18 strains,12 strains of yeast,13 strains of Trichophyton mentagrophytes.4.The ethanol extract of Gelsemium effect was stronger than that of water extract of gelsemium.The 43 strains of fungi,gelsemium water inhibiting 21 strains of fungi 100%,Ge water extract of MIC50<1.56?g/ml,MIC90=12.50 ?g/ml;GE alcohol inhibiting against 37 strains of fungi 100%,alcohol extract of Gelsemium MIC50<1.56?g/ml,MIC90=6.25 ?g/ml.5.The extraction part gelsemium ethanol extracts of petroleum ether fraction,ethyl acetate,n-butanol extract and water extract had good antifungal effect,the effect of petroleum ether extract and ethyl acetate extract is stronger than the other two parts.The 43 strains of fungi,petroleum ether extract of Gelsemium alcohol extract reached 100%inhibition of 28 strains of fungi,MIC50<0.156 ?g/ml,MIC90=20 ?g/ml;inhibition of ethyl acetate ethanol extract of Gelsemium 27 fungal strains 100%,MIC50<0.156 ?g/ml,MIC90=40?g/ml;inhibition of n-butanol parts of ethanol extract of Gelsemium on the 24 strains of fungi 100%,MIC50=0.625 ?g/ml,MIC90=10 ?g/ml;inhibition of the ethanol extract of Gelsemium water extract against 18 strains of fungi 100%,MICso<0.156 ?g/ml,MIC90=40 ?g/ml.6.For Trichophyton rubrum,antifungal effects of alcohol extract and the gelsemium extract showed strong in vitro antifungal effect of ethyl acetate extract of Gelsemium alcohol extract in vitro for best;yeast,antifungal effects of alcohol extract and the gelsemium extract showed strong in vitro,the strongest antifungal activity in vitro parts of petroleum ether and ethyl alcohol extract of gelsemium;for Trichophyton mentagrophytes,antifungal effects of alcohol extract and the gelsemium extract showed strong in vitro in vitro,including petroleum ether extract and ethyl acetate extract had the strongest antifungal effect;for Microsporum canis,gelsemium alcohol extract and extracts have strong in vitro antifungal effects on the in vitro antifungal,the petroleum ether extract of Gelsemium alcohol extract had the best effect;for Epidermophyton fungi,alcohol extraction and the gelsemium extract had strong Antifungal activity in vitro,including in vitro antifungal petroleum ether fraction of alcohol extract of Gelsemium had the best effect;for Candida albicans,gelsemium alcohol extract and extracts were 100%in vitro antifungal inhibition.7.With naftifine,gelsemium ethanol extract and its extracts for Trichophyton and Candida albicans showed antifungal effect than positive control drug naftifine better in vitro.Compared with fluconazole,gelsemium ethanol extract and its extracts for Trichophyton rubrum,yeast,Trichophyton and Candida showed better than positive control drug fluconazole antifungal effect in vitro.8.N-butanol gelsemium alcohol extract has a special concentration range 2.5-80 ?g/ml of No.14 and No.43 yeast.9.Ge ethanol extract,ethyl acetate and water in parts of Gelsemium alcohol extract of Gelsemium ethanol extract n-butanol extract and alcohol extract of Gelsemium Aspergillus anti fungal effect is not obvious.Gelsemine alcohol extracts of petroleum ether fraction are clear in vitro antifungal effects of Aspergillus,MIC50=4800 ?g/ml,MIC90=9600 ?g/ml.Anti fungal effect in vitro of petroleum ether extract of Gelsemium alcohol extract is significantly weaker than the control drug of itraconazole MIC90=4 ?g/ml.Conclusion The ethanol extract and the extraction of Gelsemium components,have anti superficial fungal strong function in vitro,and anti Aspergillus gelsemium ethanol extract and its extracts in vitro MIC50 and MIC90 value is too large,no worth in clinical application.At the same time,alkaloids have a special concentration range can inhibit the growth of yeast in yeast,which is worth further study.
