| Objective: To explore the application of modern immunohistochemical technique,Tianjing busui huoxuehuatan KaiqiaoFang on rat model of vascular dementia efficacy and mechanism of action.Method:1 by selecting SPF grade half male and half female body weight 220 + 20 g Wistar totally 60 rats,were randomly divided into model,positive control drug donepezil group,sham operation group,selection of Tianjing busui,Bushenhuoxuehuatan KaiqiaoFang new green particles in the low dose group,middle dose group and high dose a total of 6 groups,10 rats each group.The rat model of vascular dementia was modified by double vessel occlusion(2VO).2 step-down test: vascular dementia behavioral changes in rats with step-down test,traditional Chinese medicine to detect vascular dementia rats after Taking Tianjing busui,Bushenhuoxuehuatan resuscitation on the ability of learning and memory effect.3 immunohistochemical method was used to investigate the expression of Bcl-2 gene in hippocampus of rats with vascular dementia CA1 area.Result:1 Platform experimental results show that the positive control drug Aricept group and Tianjing busui,promoting blood circulation and eliminating phlegm for resuscitation in high dose group.The experimental platform can increase the latency(P < 0.05),reduce the number of errors in the step-down test in VD rats(P < 0.05).There was no significant difference between the high dose Chinese medicine group and the control group(P > 0.05).The expression level of Bcl-2 protein 2 in the model group decreased significantly(P < 0.05);and the expression of high dose group in vascular dementia rat brain hippocampus CA1 region in Bcl-2 protein levels were significantly increased(P < 0.05),while the positive drug group in the sea area by donepezil consisting of CA1 VD rats the expression of Bcl-2 compared with the model group,there was statistical significance(P < 0.05).Compared with the high dose group and positive drug donepezil group,no difference in statistics.(P > 0.05)Conclusion: vascular dementia rats learning and memory ability can fill busui,blood and phlegm resuscitation prescription significantly increased the expression of Bcl-2 in hippocampus,thus reducing the apoptosis of brain cells in rats with vascular dementia. |
