Objective:The aim of the study was to explore the effects of oxycodone hydrochloride on differentiation and cytokines of Th1?Th2 cell of mice in vivo through the intraperitoneal injection.Methods:A total of twenty male ICR mice which were five weeks old were enrolled and randomly divided into two groups:the oxycodone hydrochloride group and normal saline group.After numbered and weighed,all mice were injected intraperitoneally solution successively for nine days at the same time everyday.The oxycodone group was injected with the oxycodone hydrochloride according to the dose of 40mg/kg.Oxycodone was diluted to lmg/ml.The normal saline group was given normal saline at the dose of 40ml/kg.In the tenth day,the mice were respectively extracted the eyeball for peripheral blood and separated the serum under the sterile conditions.Then the concentration of IFN-y and IL-4 were measured by ELISA.The mice died of acute hemorrhage.Under the sterile conditions,the spleens of these mice were taken out and weight and were made into single cell suspension.The percentage of the T helper cell subsets in the cell suspensions were meaured by flow cytometry through intracellular staining assay,after T helper cells were incubated in the presence of PMA and ionomycin and the secretion of the cytokines was inhibited by BFA for five hours.Results:There was no significant difference between the spleen index of the two groups(P>0.05).There was no significant difference between proportion of Thl and Th2 cell subsets of the oxycodone group and normal saline group(P>0.05).There was also no significant difference between the ratio of Thl/Th2 of the oxycodone group and normal saline group.However,the IFN-y serum concentration of oxycodone group was significantly lower than normal saline group(P<0.01).There was no significant difference between the IL-4 serum concentration of the oxycodone group and normal saline group(P>0.05).Conclusions:oxycodone hydrochloride through the intraperitoneal injection has no obvious effect on proportion of Thl and Th2 cell of mice in vivo.oxycodone hydrochloride does not impact the Thl and Th2 cell differentiation of mice in vivo.However,oxycodone hydrochloride can reduce the serum levels of interferon-? in mice,it could partly suppress the cellular immune function.Oxycodone hydrochloride has no influence on the serum concentration of IL-4 in mice,it does not affect cytokine secretion of Th2 cell.
