| Objective:The aim of this study was to investigate the expression of BAI-1 gene in bladder normal tissues and bladder urothelial carcinoma and its effect on neovascularization of bladder urothelial carcinoma.The expression of BAI-1 gene was correlated with clinical pathological parameters and to explore the role of BAI-1 gene in the development and progression of bladder urothelial carcinoma in order to help the diagnosis and treatment of bladder urothelial carcinoma..Methods: Reverse transcriptase-polymerase chain reaction(RT-PCR)was used to detect the m RNA levels in 20 cases of normal tissue and bladder urothelial carcinoma.Retrospective analysis of the relationship between BAI-1 gene expression and clinicopathologic factors was conducted in bladder urothelial carcinoma.Immunohistochemical SP method was used to detect the expression of BAI1 in 93 cases of myometrial invasive bladder urothelial carcinoma,39 cases of non-muscular invasive bladder carcinoma and 30 cases of normal bladder tissue.The expressions of TAMs(namely CD68 +)and microvessel density(MVD,CD34 +)in the above tissues.Western Blot was used to detect the expression of BAI-1 protein in the adjacent non-myometrial invasive bladder urothelial carcinoma and myometrial invasive bladder urothelial carcinoma.Results: 1.The expression of BAI-1 m RNA in bladder normal tissues was significantly higher than that in cancer tissues,the difference was statistically significant(p<0.05).2.The expression of BAI-1 protein in bladder normal tissues was significantly higher than that in cancer tissues,the difference was statistically significant(p<0.05).3.The expression of BAI-1 in bladder urothelial carcinoma was negatively correlated with the expression of TAMs and MVD,and the expression of TAMs and MVD in bladder urothelial carcinoma was positively correlated and the difference was statistically significant(p<0.05).4.The expression of BAI-1 in bladder urothelial carcinoma was not related to age,sex,tumor size,number,smoking and tumor metastasis,but the difference was statistically significant with the stage,grade and recurrence(p<0.05).The low expression of BAI-1 had a significant effect on recurrence-free survival in patients with bladder urothelial carcinoma(log-rank p<0.05).Conclusion: 1.The positive expression rate of BAI-1,BAI-1 m RNA and its protein in normal bladder tissue was higher than that in bladder urothelial carcinoma;and the level of BAI-1 is related to the tumor staging and grading.2.The expression of BAI-1 gene is closely related to the stage,grade and recurrence of bladder urothelial carcinoma,which suggests that the low expression of BAI-1 and the negative co-ordination of CD68 + TAMs may be related to the occurrence and development of bladder urothelial carcinoma. |
