| Objective:Hyaluronic acid(HA),allogeneic bone and mesenchymal stem cells(MSCs)had been used to promote defect regeneration independently.We hypothesis that a material consist HA hydrogel(produced by a simple freezing process),allogeneic bone and enrich marrow(consist of MSCs)can be used to promote bone reconstruction.The arm of this current study was to investigate the therapeutic efficacy of rabbit femoral defect regeneration.Method: For in vivo cell proliferation,MSCs were isolated and purified for culture and serial passage by the attachment culture method.The morphological changes of the MSC were continuously observed under the inverted microscope,and membrane antigens were detected with flow cytometry.Those cells were induced to differentiate into osteoblasts,chondrocytes and adipocytes under different induction mediums.The results were observed.The HA hydrogels were prepared and the BMSCs were grown on HA hydrogels.The biocompatibility of HA hydrogels was measured by measuring cell rate,adhesion rate and effective adhesion rate.were divided into two groups as the model of the selection of the study,respectively,in the right side of the radius and the lower part of the femur caused the same area,the remaining 36.They were randomly divided into 4 groups,which were to establish femoral defect model.And concentration in blood and bone marrow standby HA hydrogels prepared;group A was biological implant group hydrogel systems,group B was HA hydrogel implant allograft Complexes,Group C was simple implanted allograft,and group D is autologous bone implants.Postoperative X-rays by the bone and a generally specimen detection repair effect,and the sections were prepared and HE staining microscopic osteogenesis repair area.Results: BMSCs were inoculated in vitro for 24 hours.The expression of CD29,CD44,CD90,CD34,CD34,CD34,proved that the cells are mesenchymal stem cells.BMSCs induced by three kinds of induced solution,after 21 d proliferation and differentiation,respectively,alizarin red,fen red O and oil red O dyeing,proved to be three The X-ray results of the animals after the built-in materials showed that the defect area of the 4weeks group A was reduced,and the 24 weeksA Group B and group B had complete repair of bone defect,and the bone repair area of B and C groups was clear and the area of normal bone was clear.The repair area did not reach the complete result.The results were consistent with the X-ray results.The HE staining showed 4weeks,A and D groups were superior to those of group B,group C,24 weeks A and D group,and the group A and group D were treated with unilateral periosteal formation in group A and group D,and the only group of 8weeks A group had intact periosteal continuity normal bone no difference,not Normal bone and / boundaries,and B,C is poor repair.Conclusion: BMSCs with high purity,good activity and stable phenotype,which can be obtained by whole bone marrow adherence method,and the ability of multi-directional differentiation can be obtained.The HA hydrogel prepared by freeze-thawing method can be used as a kind of Non-cytotoxic threedimensional cell scaffold material,and have a certain resistance to degradation;the lower part of the femur can be used as a choice of bone defect modeling,concentrated bone marrow-HA hydrogel-allogeneic bone synthesis of biological hydrogel system for bone defects Repair has a positive effect,to promote early tissue regeneration and vascular recanalization,and simple and quick operation,is a clinical treatment of bone defects in the new technology. |
