Background and Objective:Coronary atherosclerosis complicated by plaque disruption and thrombosis is a critical event in acute myocardial infarction(AMI)and stroke,the major causes of cardiovascular death.But the mechanism of atherosclerosis is very complex,and now is still not fully clear.Nowadays,most of the atherosclerotic and unstable plaque studies have focused on the effects of blood lipids,blood glucose,and blood pressure on plaque progression in the environment of human body.But little is known about the contribution of the local vulnerable plaque to the development of systematic atherosclerotic lesion.We test the hypothesis whether the local vulnerable plaque triggered by mechanical injury can promote or induce the development of atherosclerosis in other artery vessel bed distal from the target site by the secretion of certain substances.Thence this experiment sought to establish an AS animal model.Methods:Rabbit AS animal model :The total of 20 rabbits were randomly divided into three groups.Eight rabbits in the group A were fed with Western diet and subsequently had been undergone balloon injury in one carotid artery.Eight rabbits in the group B were only fed with Western diet,and four rabbits in the group C were fed with normal chow.After twelve weeks,all the rabbits were sacrificed,and specimen of bilateral carotid arter and abdominal aorta were collected for the analysis of the development of the atherosclerosis.At the same time,the serum lipid levels were assayed.Result:There were significant atherosclerotic plaques in the bilateral carotid and abdominal aorta of the group A(plaque area / vascular lumen area respectively:0.67 ± 0.01,0.48 ± 0.02,0.33± 0.05),while the group B and the group C was no obvious atherosclerotic plaque formation,the average intimal thickness was no significant difference,compared to the difference Statistically significant(p <0.05).The results of biochemical tests showed that the blood lipid were significantly increased in rabbits fed with high fat diet,and the difference was statistically significant(p <0.05)compared with the group C fed with normal diet..Conclusion:The use of balloon-damaged rabbit carotid artery combined with high-fat feeding method can effectively form a significant atherosclerotic plaque,and the formation of unstable atherosclerotic plaque can indeed affect other peripheral atherosclerotic plaque through the blood circulation.The experimental results may provide a new target of the treatment and prevention of atherosclerosis.
