Effect Of MiRNA-506 On Proliferation,Migration And Invasion Of Epithelial Ovarian Cancer Cells

Objective:To investigate the effects of miRNA-506 on proliferation,migration and invasion of epithelial ovarian cancer cells.Method:1.qRT-PCR was used to detect the expression of miRNA-506 in epithelial ovarian cancer cell lines A2780,ES-2,Ovar-3 and SK-OV-3,and then the epithelial ovarian cancer cell lines with relative high expression and relative low expression of miRNA-506 were screened out.2.The epithelial ovarian cancer cell lines with relative high expression of miRNA-506 were transfected with miRNA-506 inhibitor,and the epithelial ovarian cancer cell lines with relativerelative low expression of miRNA-506 were transfected with miRNA-506 mimics.3.The effect of miRNA-506 on cell proliferation of epithelial ovarian cancer cells were determined by CCK8.4.The effect of miRNA-506 on cell migration and invasion of epithelial ovarian cancer cells were determined by Transwell assay,respectively.Results:1.In epithelial ovarian cancer cell lines A2780,ES-2,Ovar-3 and SK-OV-3,the expression level of miRNA-506 in cell line A2780 was the highest,and in cell line SK-OV-3 was the lowest.2.Compared with inhibitor negative control group,the increased number of cell proliferation in EOC cell lines A2780 was significant in miRNA-506 inhibitor group(P< 0.05).Compared with mimics negative control group,the reduced number of cell proliferation in EOC cell lines SK-OV-3 was significant in miRNA-506 mimics group(P< 0.05).3.Compared with inhibitor negative control group,the increased number of migration in EOC cell lines A2780 was significant in miRNA-506 inhibitor group(P< 0.05).Compared with mimics negative control group,the reduced number of migration in EOC cell lines SK-OV-3 was significant in miRNA-506 mimics group(P< 0.05).4.Compared with inhibitor negative control group,the increased number of invasion in EOC cell lines A2780 was insignificant in miRNA-506 inhibitor group(P > 0.05).Compared with mimics negative control group,the reduced number of invasion in EOC cell lines SK-OV-3 was significant in miRNA-506 mimics group(P< 0.05).Conclusion:1.Knockdown of miRNA-506 could enhance cell proliferation of EOC cell line A2780;overexpression miRNA-506 could inhibit cell proliferation of EOC cell line SK-OV-3.MiRNA-506 can suppress cell proliferation of EOC cell.2.Knockdown of miRNA-506 could enhance migration of EOC cell line A2780;overexpression miRNA-506 could inhibit migration of EOC cell line SK-OV-3.MiRNA-506 can suppress migration of EOC cell.3.Knockdown of miRNA-506 could enhance invasion of EOC cell line A2780;overexpression miRNA-506 could inhibit invasion of EOC cell line SK-OV-3.MiRNA-506 can suppress invasion of EOC cell.
