Expression And Significance Of P-Akt/p-mTOR/PTEN Pathway In Mantle Cell Lymphoma

Objective: To detect the expression of phospho-Akt(p-Akt),phospho-m TOR(p-m TOR),and phosphatase and Tensin homology deleted chromosome ten(PTEN)in mantle Cell lymphoma(MCL),we study the correlation between p-Akt,p-m TOR,PTEN and clinical characteristic and its prognosis in MCL.Methods: 40 cases of MCL hospitalized in Zhangzhou Affiliated Hospital of Fujian Medical University from January 2005 to December 2016 were recruited in this study.30 cases of lymphoid reactive hyperplasia(LRH)were served as control.The expression of p-Akt,p-m TOR,and PTEN were detected by SP(immunohistochemistry).Data were analyzed by ?2 test,precise probability method,and Kaplan-Meier.Results :1.The expression of p-Akt in MCL was 82.61%(38/46),higher than that in LRH(40.0%(12/30)),P < 0.05;p-Akt was positively correlated with Ann-Arbor stage and Ki-67 in MCL,P <0.05.There were no correlation between p-Akt and age,gender,LDH,extranodal and bone marrow involve,with leukemia,P>0.05;Overall survival(OS)in p-Akt positive expression was shorter than that in p-Akt negative(36.07 months vs 76.40 months),P <0.05.2.The expression of p-m TOR in MCL was 89.13%(41/46),higher than that in LRH(6.67%(14/30)),P<0.05;There were no correlation between p-m TOR and age,gender,LDH,extranodal and bone marrow involve,with leukemia,Ann-Arbor stage,Ki-67,P> 0.05;OS in p-m TOR positive expression was shorter than that in the p-m TOR negative(37.90 months vs 72.18 months),P<0.05.3.The expression of PTEN in MCL was 34.78%(16/46),lower than that in LRH(56.67%(17/30)),P <0.05;It was negatively correlated with Ki-67,P<0.05;There were no correlation between PTEN,and age,gender,LDH,extranodal and bone marrow involve,with leukemia,Ann-Arbor stage,P>0.05;OS in PTEN negative expression was shorter than that in PTEN positive(30.09 months vs61.15 months),P <0.05.4.p-Akt was correlated positively to p-m TOR,and negatively to PTEN,P<0.05;p-m TOR was negatively correlated to PTEN,P<0.05.Conclusion: The expression of p-Akt,p-m TOR was higher and PTEN was lower in MCL.Aberrant of p-Akt,p-m TOR and PTEN may be the one of the factors for poor prognosis.Phosphrylation of p-Akt / p-m TOR / PTEN pathway might be pathogenesis and this pathway could be therapeutic target for MCL.
