Objective To explore the protective effect of ozone oxidative preconditioning on rats subjected to ischemia reperfusion injury. Methods Eighteen clean grade Sprague-Dawley male 8-week-old rats weighting 250 ~ 300 g were randomly divided into three groups(n=6 each): sham operation group(group S),ischemia/reperfusion group(group I/R),ozone oxidative preconditioning group(group O3+I/R).Group O3+I/R:Before the I/R procedure, rats received five-day ozone treatments by intraperitoneal injection of ozone and oxygen mixed gas(ozone concentration 50 mg.L-1,1mg.kg-1.once a day).Hepatic ischemia/reperfusion injury was induced by clamping the branches of hepatic artery, portal vein in the median and left lateral hepatic lobes for 45 min, followed by a 3h period of reperfusion. After reperfusion, aorta abdominalis was extract and blood samples were collected for later analysis of serum aminotransferase(ALT,AST).These rats were executed and hepatic tissue samples were collected for analysis of hepatic malondialdehyde(MDA),superoxide dismutase(SOD).Results In comparison with group S,concentrations of serum ALT,AST,hepatic MDA were increased, the concentrations of hepatic SOD were decreased(P<0.01)in group I/R,the concentrations of hepatic SOD were increased in group O3+I/R.Compared with group IR,group O3+I/R decreased concentrations of serum ALT,AST and hepatic MDA,increased the concentrations of hepatic SOD were shown(P<0.01).Conclusions Ozone oxidative preconditioning could inhibite the lipid peroxidation to protect the rats against hepatic ischemia/reperfusion injury.
