Objective:To clarify the different active substances in two botanical drug raw materials,Mori Ramulus and Mori Cortex,from different plants of Morus alba,which provides scientific basis for their quality control and national standards improvement,and gives evidence for the rational use of traditional Chinese medicine of mulberry source.Methods:1.This research analyzed the different parts of Morus alba L.by HPLC,and optimized the chromatographic conditions to maximize its characterization,so as to reveal the similarities and differences in chemical composition of different medicine parts of the Morus alba L..2.Combined with the mass information provided by UPLC-QTOF/MS,and the control compounds separated before,we identified the composition of Mori Ramulus and Mori Cortex,and cleared the same and different compositions of them.3.This research qualified several main compositions by HPLC,and summarized the differences between the two parts,such as components,proportion and other information.4.The pharmacodynamics evaluation which associated with traditional functions was carried out by their extraction with representative medicinal materials of Mori Ramulus and Mori Cortex.According to the common characteristics of Mori Cortex and Mori Ramuls,the ear swelling induced by Xylene model was used as inflammatory animal models to prove the effect of anti-inflammatory,and the cough mice model was selected to determine the antiasthmatic effect of Mori Cortex.5.The correlation of the main components and different components with efficacy wasanalyed by chemical information.Results:1.The HPLC fingerprints and common models of different medicinal parts of M.alba were established.The number of common peaks obtained from the fingerprints of Mori Cortex,Mori Ramulus,Mori folium and Mori Fructus were 10,11,12 and 8,respectively.10 characteristic peaks were identified by comparison with the reference substances and accurate molecular weights determined by UPLC-QTOF/MS.2.By identifying components of Mori Cortex and Mori Ramuls based on the exact relative molecular weights,we found 147 peaks in Mori Cortex and Mori Ramuls,35 peaks were common while 55 specific peaks in Mori Cortex and 57 specific peaks in Mori Ramuls.A total of 39 compounds were identified of Mori Cortex and Mori Ramuls based on the fragmentation information by UPLC-QTOF/MS,including 16 stilbenes,four2-arylbenzofuran,four coumarins,10 flavonoids,three D-A adducts,one phenolic acid as well as one alkaloid.3.A method for the determination for 5 kinds of main components of the Mori Cortex and Mori Ramulus was established by HPLC.52 Mori Cortex and 54 Mori Ramulus were determined.The index component in Mori Cortex are Mulberroside A,oxyresveratrol,kuwanon G,kuwanon H,morusin;The index component in Mori Ramuls are Mulberroside A,oxyresveratrol,kuwanon G,cudraflavanone C,morusin.The common elements,specific composition and their content in batches of medicinal materials were clear.At the same time,analysis the rationality of the peeling process in the origin of Mori Cortex.4.In the ear swelling induced by Xylene model,we can find that the mouse ear swelling were significantly reduced in the high dose(9.6 g/kg)and low dose(2.4 g/kg)groups of Mori Cortex and the high dose(12 g/kg)group of Mori Ramulus(p<0.05 or p<0.01).In the cough mice models test,the high and low dose of Mori Cortex can effectively decrease the times of cough in mice induced by ammonia water(the latent period p<0.01,the times of cough p<0.001;the latent period p<0.05,the times of cough p<0.01).However,there were no significant differences between the Mori Ramulus dose and the model group.5.Mori Cortex and Mori Ramuls all have anti-inflammatory effect,it may be that stilbenes(Mulberroside A)together with flavonoids(kuwanon G,kuwanon H,Ccudraflavanone C,morusin etc.)play a part in that effect.The antitussive effect of MoriCortex may be related to the high content of Mulberroside A.6.The chemical constituents,pharmacological activities and the quality evaluation of Mori Cortex and Mori Ramulus were also summed up.Conclusion:This research clarifies the similarity and difference between Mori Cortex and Mori Ramulus by qualitative and quantitative analysis of the multiple batches of medicine.Mulberroside A,oxyresveratrol,kuwanon G and morusin are the common component of Mori Cortex and Mori Ramulus,but there is a significant different in content and proportion.Kuwanon H is the unique component in Mori Cortex,while cudraflavanone C in Mori Ramulus.In addition,analysis the rationality of the peeling process in the origin of Mori Cortex.Pharmacodynamic experiments showed that both the Mori Cortex and Mori Ramulus had anti-inflammatory effects,it may be the result of interaction of stilbenes and flavonoidsit,and the antitussive effect of Mori Cortex may be related to the high content of Mulberroside A.The research results could be used for providing reference data and advice for the quality control of Mori Cortex and Mori Ramulus,as well as for the improvement of the national standard.
