Objective:this research was to detect the expression of ARID1 A,Androgen receptors in endometrial hyperplasia and its correlation with estrogen receptor and progesterone receptor.In order to understand ARID1 A and Androgen receptors with correlation between the development of endometrial hyperplasia,understand the etiology and pathogenesis of endometrial hyperplasia,explore a new idea of treatment for endometrial hyperplasia lay the theoretical basis for the prevention of and reduce endometrial cancer.Methods:132 cases of endometrial hyperplasia tissues(40 cases of simple hyperplasia,complex hyperplasia in 40 cases,52 cases of atypical hyperplasia),28 cases of proliferative endometrium were obtained from January 2013 to June 2016 at Department of Pathology Shanxi Province Department of maternal and child health hospital.All specimens were confirmed by pathology,not receiving hormone therapy within 3months.The expression of ARID1 A,ER,PR and AR in normal endometrium and endometrial proliferative lesions were detected by SP(streptavidin,peroxidase-biotin,SP).The results were observed under optical microscope and the results of immunohistochemistry were determined.SPSS21.0 statistical software package was used for statistical analysis.The expression of ARID1 A,ER,PR and AR in different endometrial tissues was examined by x2 test.The correlation between ARID1 A,AR and ER,PR expression was analyzed by Spearman.Results:1.The expression of ER,PR,AR common in endometrial tissues,the positive expression of normal endometrium group,simple hyperplasia group,complex hyperplasia group,atypical hyperplasia group ER,PR rate,no statistically significant difference between the groups(P=0.143,P=0.496);the expression rate of AR in atypical hyperplasia group was the lowest,the difference was statistically significant compared with other groups(P=0.018),proliferative endometrium,simple hyperplasia,complex hyperplasia group group,the positive expression rate of AR,the difference was not statistically significant(P>0.05)2.ARID1 A gene was expressioned both in endometrial glandular epithelial and stromal cell nucleus,with the development of endometrial hyperplasia and deletion of the ARID1 A gene expression rate increased gradually in atypical endometrial groups,ARID1 A expression rate was 19.2%,compared with the other groups,the difference was statistically significant(x2=11.58 P=0.004).There was no significant difference among other groups(P>0.05).3.There was no significant correlation between ARID1 A and ER,PR and AR in endometrial hyperplasia(rs=-0.066,P=0.455;rs= 0.001,P= 0.987;rs= 0.114,P= 0.191).The expression of AR was positively correlated with the expression of ER and PR(rs=0.176,P=0.043;rs=,0.195,P= 0.025).Conclusion1.ARID1 A gene mutation and deletion is an early event in the process of malignant transformation of endometrial hyperplasia.2.the occurrence and development of androgen and its receptor may be involved in endometrial hyperplasia.3.For some patients with AR positive endometrial hyperplasia,combined with reduced androgen therapy,may become a new strategy for the treatment of endometrial hyperplasia.
