Objective:To examine the effect of allogenic mesenchymal stem cells transplantation on the level of IL-22 and MMP-3 of serum and synovium of rats with collagen induced Arthritis.Methods:Rat CIA model was established by immunizing SD rats with type ? collagen for two times and48 SD rats were randomly divided into three groups: Normal group,CIA control group,CIA test group.MSCs that the phenotypes were assessed by flow cytometry were purified and cultured from the bone marrow of SD rats,and then others were transplanted to the rats of CIA test group in the way of 1×107/kg.The level of IL-22 and MMP-3 of serum and ankle joint were measured by ELISA and immunhistochemistry.Synovial fibroblasts were isolated and cultured and then treated with recombinant rat IL-22(rr IL-22),and the MMP-3 mRNA and protein concentration of synovial fibroblast were measured by RT-PCR and Western blotting.Results:After MSC transplantation,CIA rat arthritis index,X-ray,HE staining showed that joint damage significantly reduced compared with the control group;the ELISA results showed that the expression of MMP-3 and IL-22 of CIA control group was highest,and compared to the normal control group(125.79±9.12 VS 102.00±7.63 ng/ml,P < 0.05),(292.35±31.23 VS 257.27±13.99 ng/ml.P < 0.05)and CIA group(125.79±9.12 VS 97.94 ±9.50 ng/ml,P < 0.05),(292.35±31.23 VS 262.16±22.02 ng/ml,P < 0.05)the difference was statistically significant(P < 0.05),Normal group and CIA experimental group had no difference statistical significance;The result of immunhistochemistry was in accordance with ELISA.In RA synovial fibroblasts treated with rrIL-22,the expression of RANKL mRNA and protein was increased in a dose-dependent manner.Conclusion:These results shows that IL-22 and MMP-3 is involved in the pathogenesis of Rheumatoid arthritis.IL-22 up-regulates MMP-3 expression in RA synovial fibroblasts.Transplantation of MSCs ameliorates RA through the modulation of the expression of IL-22 which regulates the expression of MMP-3.
