Screening Of Perillaldehyde Type Perilla Germplasm And Study On The Change Rules Of Perillaldehyde Content | Posted on:2018-01-18 | Degree:Master | Type:Thesis | Country:China | Candidate:C W Zhang | Full Text:PDF | GTID:2334330536974318 | Subject:Pharmaceutical | Abstract/Summary: | PDF Full Text Request | Objective:Perillaldehyde type is qualified for Perillae Folium source in “Chinese Pharmacopoeia”.but the investigation of Perilla leaf resources showed that most of China’s existing perilla leaf medicines are perillaketone type in which perillaketone has toxic side effects.This situation seriously affects the effectiveness and safety of perilla leaf medicinal materials.So it is important that perillaldehyde type germplasm resources selecting should be carried out to provide the breeding of fine varieties of perillaldehyde type of Perilla.And by planting perillaldehyde type germplasm and determine perillaldehyde content in plants growing periods to provide some guidance for the production practice and Perilla planting and harvesting of Perilla leaf.Methods and Results:Preliminary screening of Perilla fruit morphology by observation and comparison,5characteristics are important to describe Perilla fruit: surface color,ridge height,hardness,diameter and 100 grains weight.55 Perilla fruits were described and clustered based on the data.6 categories are divided and each category was sorted out subspecies type and essential oil chemotype further after planting and volatile determination.It is got the conclusion that perillaldehyde type appears only in the fruits with small corn(the average size less than 1.5mm,100 grains weight less than 0.16g),yellow brown or dark brown,hard,high ridge features.But in this type only 30-50% germplasm are Perillaldehyde type and all of them are P.frutescens var.crispa(Thunb.)Hand.-Mazz.To establish a rapid determination method of perillaldehyde content HPLC The method of perillaldehyde determiation was established for rapid determination perillaldehyde of Perilla leaf and fruit by reference “Japanese Pharmacopoeia”.Validation of Perillaldehyde type germplasm screening by the fruit shape and fruit Perillaldehyde existing Perillaldehyde perilla fruit type(small yellow brown or dark brown,hard,high ridge)was confirmed by more fruit samples.When combining with fruit perillaldehyde determination,identification rate of perillaldehyde type germplasm can reach more than 80%.Study on the changes of perillaldehyde content and the evaluation of perillaldehyde germplasm 9 perillaldehyde type germplasms were planted in IMPLAD experimental field in Beijing.Leaf samples of different growth development period taking into account one day picking time and plant parts were collected and measured perillaldehyde content.The content difference of different perillaldehyde germplasm is great.High content is from samples collecting date before September 21 stthat is the before flowering period.The content of different plant parts is: top leaves > middle leaf > bottom leaves,and in deciduous period tending to be similar.The different picking time is with little influence on content.Conclusion:This paper established a method to screen perillaldehyde type germplasm combining with perilla fruit shape and perillaldehyde rapid detection feasibility and reliability.Through the large amount perillaldehyde content data analysis of multiple type germplasm perillaldehyde in the planting process,it is showed that the main influencing factors of perillaldehyde content is germplasm,followed by collection date and collection part,little effect of different picking time of the day.Most of the germplasm before heading or flowering perillaldehyde content was the highest.It is possible that blossom affects perillaldehyde content accumulation.The content of the top leaves was higher thanother parts.4 perillaldehyde germplasms with high content were selected.The result of this pater will put forward the appropriate collection time in the production of Perilla leaf to ensure the Perillae Folium herb quality. | Keywords/Search Tags: | Perilla, Fruit morphology, Perillaldehyde type screening, Perillaldehyde content change rule | PDF Full Text Request | Related items |
