Objective: To prepar intermittent hypoxia rats model,and to detect the amount of glucose transporter 2(GLUT-2),glucokinase(GCK)protein expression in liver cells,and determination of the degree of insulin resistance,to explore the related mechanism between the expression of GLUT-2 and GCK in intermittent hypoxia and the occurrence of insulin resistance.Methods: 24 healthy male SD rats were randomly divided into control group(NC group),intermittent hypoxia 4 weeks group(IH4)and intermittent hypoxia for 8 weeks group(IH8),with a total of 8 rats in each group.The model groups were given the preset ventilation mode exposure to intermittent hypoxia(9:00am-5:00pm),the control group was given intermittent compressed air at exposure time,the normal ventilation rest time.At the end of the experiment,fasting blood glucose and fasting insulin were measured in each group,and insulin resistance index(HOMA-IR)and insulin sensitivity index(ISI)were used to evaluate the degree of insulin resistance.Immunohistochemical staining was used to observe the changes of GLUT-2 and GCK protein expression in liver cells,and the average gray value was determined by quantitative analysis of protein expression,at last integrate data and statistical analysis.Results: 1.Compared with the NC group,the fasting blood glucose,fasting insulin,HOMAIR in model group were increased,ISI were decreased,and with the extension of exposure time,the above changes were more obvious,the difference was statistically significant(P <0.05);2.The expression of GLUT-2 and GCK protein in liver cells decreased under intermittent hypoxia,and the changes were more significant in the 8 weeks group than that in the 4 weeks group(P <0.05);3.Pearson correlation analysis showed that GLUT-2 and GCK average gray values were positively correlated with HOMA-IR(r=0.894,r=0.869,P<0.05),and negatively correlated with ISI(r=0.886,r=0.906,P<0.05).Conclusion: 1.Groups of rats exposed to intermittent hypoxia the fasting blood glucose,fasting insulin and HOMA-IR increased,ISI decreased,the body showed signs of glucose metabolism disorder and insulin resistance,and with the exposure time prolonged the change will be more obvious;2.The expression of GLUT-2 and GCK protein was down regulated under intermittent hypoxia,and the gray value of the two proteins were correlated with HOMA-IR;3.The expression of GLUT-2 and GCK protein in liver cells participate in the occurrence of hepatic insulin resistance in intermittent hypoxia.
