Objective:To explore effect of edaravone intervention on serum NSE level of neonatal rats with hyperbilirubinemia.Methods:A total of 72 7-day-old SD rats were randomly divided into H,E and C groups: H group was hyperbilirubinemia group(n=24),E group was edaravone intervention group(n=24)and C group was normal control group(n=24).According to the different time point,each group was randomly divided into 3 subgroups: 12 h Group,24 h group and 48 h group,8 in each subgroup.The neonatal rats in group H and group E were injected intraperitoneally with bilirubin as a dose of 100?g/g,only 1 times;neonatal rats in group C were injected intraperitoneally with normal saline as a dose of 100?g/g,only 1 times;after establishing animal model of hyperbilirubinemia,neonatal rats in group E was given edaravone intervention immediately(2mg/kg/times,once a day).To observe whether there was abnormal behavior in neonatal rats,such as convulsive,reduced activity,etc.And to take blood and brain tissue at each time point.The pathological changes of brain tissue of neonatal rats was observed by using hematoxylin-eosin staining method.Serum NSE was measured by enzyme-linked immunosorbent assay(ELISA).Results:1.There were no obvious abnormalities in behavior of neonatal SD rats in H,E and C3 group before experiment.After model established,rats in group H and group E had abnormal behavior,such as convulsive,reduced activity,etc.But group E was lighter in behavior abnormalities than group H.The neonatal SD rats in C group did not show obvious abnormal behavior in whole experiment.2.HE staining results: the cells in the hippocampus in C group were arranged neatly,the morphology and structure of the cells were intact,and there was no obvious edema;the cells in the hippocampus in H group were slightly disordered,with obvious edema and obvious inflammatory cell infiltration;there were a few nerve cell edema in brain tissue of E group,but at each time point the pathological changes of brain tissue of E group were lighter than that in H group.3.The levels of serum NSE in group H were higher than that in group C at each time point,the difference was statistically significant(P <0.05);At each time point,the level of serum NSE in group E was lower than that in group H,but it was still higher than that in group C,the difference was statistically significant(P <0.05).Conclusion:Serum NSE level of neonatal rats with hyperbilirubinemia increased,edaravone intervention could reduce level of serum NSE,showed that edaravone has neuroprotective effect on brain injury in neonatal rats with hyperbilirubinemia.
