| Objective:To explore effect of Yishen capsule on the rat renal tubular epithelial cell autophagy mechanism under the condition of high glucose stimulation.Methods:Renal tubular epithelial cells are divided into 6 groups:(1)normal glucose levels group(NG):the concentration of glucose is 5.5mmol/l;(2)high glucose levels group(HG):the concentration of glucose is 30mmol/l;(3)low-dose of Yishen capsule treatment group(YL):0.625g/kg/d;(4)medium-dose of Yishen capsule treatment group(YM):1.25g/kg/d;(5)high-dose of Yishen capsule treatment group(YH):2.5 g/kg/d;(6)Cozaar group(LT):20 mg/kg/d,Groups of cells were cultivated for 24 hours,respectively.To observe the autophagosome in the renal tubular epithelial cells of each group under the transmission electron microscope,and The expression of LC3 protein and P62 protein in the renal tubular epithelial cells were detected by Western blot.Results:(1)The autophagosome quantity in the renal tubular epithelial cells of each group under the transmission electron microscope:compared with NG group,autophagosome number of HG group was decreased significantly(P< 0.05);Compared with HG group,YL,YM,YH and LT group autophagosome quantity were increased(P< 0.05),but no statistical difference was found between YH and LT group.(2)The expression of LC3 protein and P62 protein in the renal tubular epithelial cells of each group:compared with NG group,the expression of LC3 in HG group was obviously decreased(P<0.05),p62 expression increased(P<0.05);compared with HG group,the expression of LC3 in YL,YM,YH,LT group increased(P<0.05),and p62 expression decreased(P<0.05),but no statistical difference was found between YH and LT group.Conclusion:High glucose environment can inhibit autophagy function,however,Yishen capsule can activate autophagy functions and autophagy effect is related to drug concentration,Which shows that Yishen capsule has protective effect on renal epithelial cells. |
