| Objective : To observe the Effect of Yishen Capsule on expression of SOCS-3 and TGF-beta 1 in renal tubular epithelial cells of the rats with diabetic nephropathy(DN) for further exploring progression of Yishen Capsule in treatment of diabetic nephropathy.Methods : Normal rat proximal tubular epithelial(NRK52E) cells were cultured in RPMI-1640 medium, then collected rats’ blood and filtered to prepare medicated serum.Randomly divided into 7 groups: normal control group, isotonic group,high glucose group, low-dose of Yishen Capsule treatment group(1.5g/kg/d),medium-dose Yishen Capsule treatment group(3.0g/kg/ d), high-dose Yishen Capsule treatment group(6.0g/kg/d) and benazepril treatment group(30mg/kg/d). Expression of SOCS-3 and TGF-β1 in rat proximal tubular epithelial cells were detected by Western blot, and the expression of SOCS-3m RNA were detected by blot rt-PCR, and immunofluorescence staining was used to determine expression and distribution conditions of TGF-β1 and SOCS-3.Results : Real-time PCR was used to detect the results suggest that expression of SOCS-3m RNA was significantly increased(p<0.05) under the condition of high glucose stimulation;compared with high glucose group,Yishen Capsule treatment groups’ expression of SOCS-3m RNA had further increased. While western blot and immunofluorescence staining method was used to detect the results suggest that expression of SOCS-3 and TGF beta 1 were significantly increased(p<0.05) under the condition of high glucose stimulation;compared with high glucose group,Yishen Capsule treatment groups’ expression of SOCS-3m RNA and protein had further increased.Conclusion : Yishen Capsule may ameliorate the tubular epithelial-mesenchymal transdifferentiation of diabetic rats by regulating overexpression of SOCS-3 and inhibiting overexpression of TGF-β1. |
