Effect Of Bifid Triple Viable Capsules On The Expression Of IL-6 In Intestine And Livers Of Mice With Experimental Colitis

Ulcerative colitis(UC)is a kind of chronic,recurrent inflammatory bowel disease,of which the etiology is poorly known.The incidence of UC is increasing worldwide.At present,UC is thought to be related to infection,environment,immunity,intestinal mucosal barrier dysfunction,intestinal microecology and heredity and other factors.Although UC lesions mainly involve the intestinal tract,various forms of parenteral manifestation are common,with liver as one of the most common organs.The current studies speculate that UC increases the intestinal mucosal barrier permeability,and thus leads to the translocation of intestinal flora into the bloodstream,causing portal vein bacteremia.Subsequently,intrahepatic inflammatory mediators(such as IL-6)are activated,which results in liver and gallbladder disease.Bifid triple viable capsule(BTVC),which is composed of the Lactobacillus,Bifidobacterium and Enterococcus faecalis,can form a biological barrier in the intestinal surface to reduce the permeability of intestinal mucosa.The reduced permeability then prevents translocation of bacteria and decreases endotoxin production and absorption,thereby inhibiting the release of inflammatory factors.In order to explore the effect of BTVC on the the disease activity index(DAI)and expression of IL-6 of the colon and liver in the DSS-induced colitis model,45 BALB/c mice were randomly divided into 3 groups,including normal group,DSS group,and DSS+BTVC group.Ten days later,blood,colon and liver tissues were collected.The length of the colon was measured and the morphological and histopathological changes were observed.ELISA was used to detect the expression of IL-6 in the colon and liver tissue.The results showed that DSS group significantly increased the disease activity index and the level of IL-6 in colon and liver tissues.On the other hand,the application of BTVC significantly decreased DAI and the level of IL-6 in colon and liver tissues.The present study confirmed that BTVC has a therapeutic effect on DSS-induced colitis and reduces the damage of liver.This study also shows that BTVC can reduce the expression of IL-16 in the colon and liver in DSS-induced colitis model.Our study provides a novel therapeutic strategy for UC-related hepatobiliary lesions.
