ObjectiveTo study antigenicity of cross-antigens of M.smegmatis(MS)and M.tuberculosis(MTB).MethodsBacteria antigens from M.smegmatis and M.tuberculosis were extracted by ultrasonication and vigorous stir,as well as late secretory protein gathered by myself.Homologous antigens reacted with both MS serum and MTB serum were analyzed by Western blot.Antigens were mixed with incomplete freund's adjuvant and then injected into mice muscle.IgG in serum was detected with ELISA 7 days later,and then 3 kinds of cytokines of spleen lymphocytes stimulated by both of MS and MTB antigens were determined with flow cytometry.meanwhile,the level of antigens-specific lymphocytes were detected with ELISPOTResultsSome homologous antigens between 10 KD to100KD of MS and MTB can be found from the results of western blot.Potent humoral immune responses and cellular immunity against both of MS and MTB can be induced by these cross-antigens after sensitization by either MS or MTB antigens.After stimulated by MS and MTB antigens,IL-2 and IFN-? secreted by CD4+ and CD8+T cells were much increased in immune body compared with cells without sensitization and stimulated by antigens of bacteria Brucella.ConclusionHomologous antigens between M.smegmatis and M.tuberculosis can effectively promote body's specific immune reactions to the infection of M.tuberculosis,which provided a scientific evidence for the development of tuberculosis vaccines.
