Tuberculosis(TB)is one of an ancient disease caused by Mycobacterium tuberculosis(Mtb).TB can spread with a concealment and continuity way but cannot easily been detected,it's characteristic of the high drug resistance of mycobacterium tuberculosis.It hunter in the cell of animal can escape from immune cells attack.Also the important characteristic of TB is that TB can survive for a long time in animals.Mycobacterium tuberculosis can stay in the human body for years without causing disease,a state referred to latent tuberculosis.Following with the immune system function decreasing or injured,TB can outbreak anytime and threaten human health in the future.The first step is recognition of mycobacteria as invading pathogens,followed by activation of innate host defense responses and the subsequent initiation of adaptive immune responses.To elucidate the pathogenesis of tuberculosis and develop new treat for TB is very significance.The outermost components of the M.tuberculosis whole cell lysis,predominately lipids and carbohydrates,are the first to contact with host molecular constituents and play a major role in facilitating host cell to recognition and modulation of host responses.TB early interactions events with the host,may determine the fate of the bacteria.It's key to Mycobacterium tuberculosis remain dormant in the human body for years without causing disease,a state referred to as latent tuberculosis.Purpose: To elucidating the role of M.smegmatis whole cell lysis for impacting on the bacteria removal rate and defending the inter-reaction of host immune.Methods: We built in vivo models,for the fatal virulence of TB,we chose M.Smegmatis mc2155,which has both close genetic background and similar immune response in all the infection events.we also use Escherichia coli as a control bacterial which is most easy contact but less possible to cause pulmonary infection.We check the pathology state of the infection site of lung and the change in systemic immune.In this part of work we will mainly use WCL,to check the role it acts for bacterial resistance during infection.check the pathology state of the infection site of lung and the change in systemic immune.Discuss M.Smegmatis WCL and E.coil WCL on the host immune system regulation.Results:(1)In this experiment show that M.Smegmatis in mice clean rate lower than E.coli,significant differences(P<0.05).Use M.Smegmatis WCL interference of E.coli,E.coli in the host clean rate turn to slow,M.Smegmatis WCL adjust the bacterial clearance rate,significant differences(P<0.05).(2)HE staining immune pathology results show that E.coli immune cells is higher than M.Smegmatis and M.Smegmatis WCL.Interference of E.coli.WCL causing immune cells aggregation also higher than M.Smegmatis WCL.(3)ELISA detection M.Smegmatis infection in mice lung,pro-inflammatory factor secretion of IFN-? lower than E.coli infection group,inflammation suppression factor secretion of IL-10 higher than that of E.coli infection group,M.Smegmatis WCL interference of E.coli caused by lung pro-inflammatory factor secretion of IFN-? clean rate reduced,lung inflammation suppression factor secretion of IL-10 rise;The same result between EW and SW.(4)CBA detection of humoral immunity,M.Smegmatis infection after host the pro-inflammatory Th1 factor IFN-?,IL-17 and TNF lower than E.coli infection group,Th2 factor IL–4,however,suppression of inflammation factor secretion of IL-10 higher than that E.coli infection group.M.Smegmatis WCL interference of E.coli caused by the host inflammation suppression factor secretion of IL–4 increase;Comparing the M.Smegmatis WCL and E.coli WCL change host's immune system,also can make host immune attack effectively,provide a new research direction for tuberculosis immunity mechanism study.Conclusion: M.Smegmatis WCL adjust the bacterial clearance rate and regulating the host immune system to identify the key influencing factors.M.Smegmatis can survive for a long time in the host,the key to escape from immune attack,so that the bacteria can survive in the host for the long time to provide favorable conditions and provide a new research direction for tuberculosis immunity mechanism study.
