ObjectThe volume of human hippocampus were measurement by manually automatic,Freesurfer and VBM.To investigate the correlation and consistency of two kinds of automatic measurement with manual measurement in order to select the best way to replace the manual segmentation of the hippocampus volume,for clinical diagnosis and differential brain volume anomalies a fast and accurate way.Try to establish the MRI measurement standard of the total,head,body,tail volume of hippocampal and analysis of AD in the course of each gender,age differences and law,to provide basis for early diagnosis of pathological changes of hippocampal structure,combined with the early clinical manifestation of AD to replenish the correlation by AD and hippocampal volume.MethodsMedical examination center of First Affiliated Hospital of Chongqing Medical University collected 321 subjects,excluding brain diseases and mental disorders underwent conventional MRI scans,the subjects were adults,obtaining the whole brain T1 image,using the method outlined manually measuring head on the left,the anteroposterior and vertical axis length of hippocampus amygdala,hippocampus,hippocampal volume,high volume.According to 18-29 years group,30-39 years group,40-49 years group,50-59 years group,60-69 years old age group,and the differences in the above data hippocampus between the F group and randomly selected from trend;manual measurement in youth(20-40 years old)of 67 subjects,aged(60-80 years old)in 75 cases,get in the volume of the whole brain MRI scan were introduced into Freesurfer and VBM8,and the gender differences in each age group,choose the optimal measurement method.To eference AD process,collected 90 cases of subjects with brain MRI images,divided into normal control group(NC group),mild cognitive impairment,Alzheimer's disease(MCI)group(AD)for manual measurement and analysis,head MRI scan after measuring subjects on the cranial cavity diameter,cranial cavity diameter,anteroposterior diameter of cranial cavity left and right hippocampal volume and hippocampal head,body and tail of three-dimensional segmentation and measurement of volume,and the subjects' mini mental state examination(Mini-Mental State,examination,MMSE),ADL(Activity of Daily Living Scale,ADL),the clinical dementia rating scale(Clinical dementia rating scale,CDR),the Montreal cognitive assessment(Montreal Cognitive,Assessment,MoCA),Hamilton Depression Scale(HAMD)assessment results.Comparison of AD,MCI,NC three groups of hippocampus volume data age,gender and left and right measurement differences,the volume of the nerve and the scale of the consistency of the analysis.ResultsNormal adults use manual delineation measurement results of total volume of the hippocampus,head of hippocampus volume,volume,volume of hippocampal tail 18-29 age group gender differences were p=(0.132,0.172,0.276,0.087),30-39 group(0.152,p= 0.261,0.829,0.072),40-49 group(0.172,0.092,0.069 p= 0.071,50-59),group p=(0.023,0.017,0.048,0.046),60-69 group(0.010,p= 0.005,0.021,0.001);linear comparison,standardization and hippocampal long axis in gender differences found only in the 40-49 and 50-59 age groups(p=0.042,0.038),and standardization long,long axis in hippocampus hippocampus 40-49 years group,50-59 years old group of women were more than men(P<0.05).The use of Freesurfer,manual delineation of normal adult and VBM8 measurement of left hippocampal volume were 3594.53 youth group,4385.72 + 377.26 mm3 + 483.11 mm3 4033.17 + 306.78 mm3;the elderly group of 3487.08 + 363.15 mm3,361.53 mm3,4023.82 + 3906.25 + 410.32 mm3;don't youth group of 3768.05 + 415.37 mm3,4483.25 mm3.18,4208.41 + 467 + 382.76 mm3 for the right hippocampus is integral;the elderly group of 3682.74 + 425.58 mm3 4165.70 + 421.56 mm3 4158.62 + 370.59 mm3.There were significant differences between the two methods of automatic measurement and manual measurement of ICC,the correlation between VBM and manual measurement was higher than that of Freesurfer,and the young group was older than that of the old group,and the right side was on the left side(P < 0.01).Healthy elderly control group,MCI group and AD group of upper and lower cranial diameter were 141.74 + 6.96 mm,144.88 + 8.06 mm 145.79 + 7.27 mm;about cranial cavity diameter were 136.93 + 5.93 mm,6.53 mm,137.18 + 138.78 + 5.12 mm;cranial anterior posterior diameters of 173.40 + 9.12 mm,172.34 mm,174.74 + 7.65 mm + 7.40.After the standardization of NC group,MCI group,AD group,the total volume of the left hippocampus were 3254.73 + 232.88mm3,3005.50 + 414.85,2540.06 + 464.33 mm3 mm3;left the sea horse volume were 1934.04 + 131.88 and 1784.03 + 272.48 mm3 + 342.19 mm3,1410.74 mm3;the left hippocampal volumes were 838.46 + 70.31 mm3,761.15 mm3 + 121.77,698.20 + 102.01 mm3;left the sea Masson volume were 482.24 + 54.32 and 460.32 + 77.10 mm3 + 82.82 mm3,431.14 mm3;the total volume of the right hippocampus were 3270.93 + 222.75 mm3,410.48 mm3,3018.62 + 2537.20 + 475.72 mm3 1934.50 + 129.01;don't mm3,1753.81 + mm3,1395.42 + 263.07 345.90 mm3 sea horse right body integral;right hippocampal volumes were 837.94 + 60.89 mm3,121.89 mm3,799.80 + 706.94 + 101.25 mm3;the sea right cauda equina volume were 498.50 + 52.94 mm3,465.01 mm3,434.84 + 86.7 + 83.22 6 mm3.In group AD,the left and right hippocampus(LHP,RHP)volume was less than NC group(P < 0.01),< MCI(P < 0.05);the left and right hippocampus volume of group MCI was less than group NC(P < 0.05).ConclusionNormal adults in(18-49)the volume of hippocampus remained stable,about measurement and sex difference is not obvious,(50-69)at the age of the subjects of the measurement values were significantly reduced,and with the increase of age is shrinking faster trend,female male right hippocampal volume>left.Freesurfer can be used in the youth group hippocampal volume measurement;VBM can be more accurate to replace the manual segmentation by measurement of hippocampal volume is Freesurfer,which shows that VBM can be better used to measure brain structure volume required for clinical disease or experiment.The volume of left and right hippocampal segments was significantly reduced in patients with AD,and the volume of hippocampus tail atrophy was not significant in MCI,but the volume of left and right hippocampus was significantly reduced.Combined with the early clinical manifestations of AD patients,it is suggested that the hippocampus is involved in the cognitive function of the human brain.
