| Gastric cancer(GC)remains one of the most common cancers and a major cause of cancer-related death worldwide.To date,tumor resection has remained the only curative therapy for gastric cancer.Some reports indicated that Cytokine induced killer cells(CIK cells)co-cultured with Dendritic cells(DC)had a high proliferation rate and enhanced cytotoxic activity.And some clinical research had been carried through about therapy for cancers in blood system or other systems with CIK cells co-cultured with DC.Objective: To evaluate the recent clinical effect and variation of immune function of chemotherapy combined with dendritic cell-cytokine induced killers(DC-CIK)on the patients with advanced gastric cancer.And the saftey was observed.The purpose of this study is to explore a new way of immunotherapy for gastric cancer.Methods: Fifty patients with advanced gastric cancer admitted in The Second Affiliated Hospital of Qingdao University(Qingdao Central Hospital)from January 2011 to December 2015 were enrolled in the study.Patients were randomly assigned in two groups.25 cases received DC-CIK combined with chemotherapy(observation group)and 25 cases at the same period received routine chemotherapy.25 in control group,received chemotherapy alone.25 in observation group,received DC-CIK combined with chemotherapy.The immune function,recent efficacy and toxicity were evaluated and compared between two groups.The percentage of T cell subsets were detected by flow cytometry,the level of cytokines was detected by enzyme-linked immunosorbent assay(ELISA).Short-term clinical efficacy was evaluated according to RECIST criteria.Measurement data were compared by t test,P<0.05 as statistically significant difference.The safety were also observed.Results: After treatment,the percentage of T lymphocyte subtypes CD4+ and CD3+ in the peripheral blood were higher in the observation group,and the level of IFN-γ 、IL-12 was higher,all P<0.05 as statistically significant difference.After treatment in control group,the percentage of T lymphocyte subtypes CD8+ T cells was increased significantly and the level of IFN-γ was reduced,P<0.05 as statistically significant difference.There were significant differences in the levels of IL-2 in the observation group and control group.While the observation group’s disease control rate is higher than control group,the recent clinical effect of the two groups had no significant difference(P>0.05).In the course of dendritic cell-cytokine induced killer cells feedback,obvious adverse reaction didn’t appear.Conclusions: cell-cytokine induced killers(DC-CIK)cell immune therapy in advanced gastric cancer has certain clinical curative effect.Chemotherapy combined with DC-CIK can improve the immune function and has little adverse reactions.DC-CIKs cell-therapy is likely to be an effective adoptive immunotherapy for advanced gastric cancer. |
