Expressions And Significance Of Autophagy Related Gene ARHI And LC3 In Cervical Cancer

Objective:Test the expression levels of the autophagy gene ARHI and LC3 in normal cervix tissues,CINII-III tissues and cervical cancer tissues respectively,and analyze the relevance of them in cervical cancer tissues and the relationshipbetween their expressions and the pathological features of cervical cancer.To discuss the role of autophagy gene ARHI and LC3 in the occurrence and development progression of cervical cancer.Methods: Examine the expression of autophagy gene ARHI and LC3 in normal cervix tissues,CINII-III tissues andcervical cancer tissues by immunohistochemical staining,analyze their expressions and the relationshipbetween their expressions and the pathological features of cervical cancerwith ?~2 test,and analyze the relevance of ARHI and LC3 in cervical cancer with Spearman mothod.Results: 1.The positive expression rate of ARHI in normal cervix group,CINII-III group and cervical cancer group were 85.0 %,73.3% and 54.0% respectively.Compared among the three groups,the difference was statistically significant(?~2=7.145,p<0.05).The positive expression rates of LC3 in normal cervix group,CINII-III group and cervical cancer group were 95.0 %,56.7% and 38.0 %,respectively.The differences were statistically significant(?~2=18.801,p<0.05);2.The expression level of ARHI and LC3 were related to clinical stage and tissue differentiation degree of cervical cancer.The differences were statistically significant(P<0.05).However,there was no significant correlation between ARHI?LC3 and the lymph node metastasis,age,SCC and surgical margin((P>0.05).3.The expression of ARHI and LC3 in cervical cancer group has a positive correlation(r=0.43,p <0.05),which indicate that ARHI?LC3 are closely related to the occurrence and developmentof cervical cancer,and both them have synergistic effect.4.There are 68 patients have been found HPV infection among the 100 patient.According to the degree of HPV infection is divided into three groups,with the increase of HPV infection,ARHI and LC3 showed a decreasing trend.The expression level of LC3 was related to the degree of HPV infiection and the differences were statistically significant(?~2=10.583,p <0.05),whereas,the expression level of ARHI was not correlated with the degree of HPV infection,the difference was not statistically significant(P>0.05).Conclusions:The decrease and loss of ARHI and LC3's expression is closely related to the occurrence and progression of cervical cancer,and they have synergistic effect to each other.The expression level of LC3 is related to the degree of HPV infection.
